Platelet Antibody Analysis by Three Different Tests

Abstract: ELISA, solid-phase assay, and LCT showed highly divergent results. Due to several limitations, the additional analysis by the "monoclonal antibody-specific immobilization of platelet antigen" (MAIPA)-assay is highly recommended.

“…False‐positive results using this assay also reportedly have occurred as a consequence of human anti‐mouse antibodies . False‐positive results also may have occurred because of low positive OD cutoff thresholds, as suggested by others . Of note, in January 2016, the cutoff OD thresholds to define positive results were increased for the LIFECODES PakPlus screen.…”

“…Alternatively, the conflicting results may be due to the different methods used for platelet antibody detection. Different platelet antibody detection assays can yield highly divergent results . The earlier study employed the Capture‐P Ready Screen and only estimated platelet‐specific glycoprotein immunization by attempting to elute HLA class I antigens with chloroquine treatment.…”

“…Such an approach has a very low sensitivity and is always inferior to a direct GPspecific test. 33,34 However, in some ITP patients with very low platelet counts, a direct test cannot be performed, and a positive indirect test may be considered helpful.…”
