Platinum Compounds: A Hope for Future Cancer Chemotherapy

Abstract: The discovery of cis-platin and its second and third generation analogues created a hope in cancer chemotherapy. Cis-platin and its second generation analogue carboplatin have been used for the treatment of some cancers from a long time. The third generation analogues have superior anti-cancer profiles for curing a few cancers. Unfortunately, certain side effects such as renal impairment, neurotoxicity and ototoxicity etc. are associated with these drugs. But, combination therapy makes these analogues more eff… Show more

“…[19][20][21] Although major advances have been made by researchers but the medical need is still largely unmet due to many factors; among which are the lack of selectivity of conventional drugs leading to toxicity, the metastatic spreading, and multi -drug resistance. [22][23][24][25][26][27] Novel and selective anticancer agents are urgently required due to problems associated with currently available anticancer drugs. The plant Euclea natalensis (A.…”

“…Age-related hearing loss, noise-induced hearing loss, and ototoxicity has been shown to occur as a result of an apoptotic process [3][4][5][6][7] . KRG extracts, or its specific saponins, were shown to have antiapoptotic and antioxidant properties in various tissues both in vitro and in vivo [8] .…”

“…Ototoxicity could lead to language and speech disabilities in young children [1][2] . The mechanism of CDDP-induced ototoxicity is apoptosis, and outer hair cells, stria vascularis, and spiral ganglion neurons are mainly affected [3][4][5][6][7] .…”

“…Cisplatin is one of the most potent antitumour drugs available for the therapeutic management of solid tumours, overian, lung, head, neck, and bladder cancers, etc. Despite wide application as a chemotherapeutic agent, cisplatin exhibits severe side effects, such as nephrotoxicity, neurotoxicity, ototoxicity, nausea, and emetogenicity, etc., which limit possibilities for gaining therapeutic benefits from dose intensification [5,6]. Thus, scientists are now engaged to explore the transition metal based complexes and other metal based complexes.…”
