Pluripotent stem cells and tolerance induction in organ transplantation

Abstract: PSC and/or their derivatives possess unique immunological properties that allow for acceptance of PSC-derived tissue with minimal host conditioning. Investigators involved either in regenerative or in transplant medicine must join their efforts with the ultimate aim of using PSC as a source of donor-specific cells that would create a protolerogenic environment to achieve tolerance in solid organ transplantation.

“…Early studies of hESC demonstrated low levels of HLA class I antigens, with a modest induction during differentiation, and absence of HLA class II 12 , 13 . Similar results have been reported for hiPSC 14 . In a recent study, including both hESC and hiPSC lines, Chen et al .…”

“…Successful PSC-based therapy will also require limiting the risk of immune response to PSC derivatives. Although PSCs display some level of immuno privilege as they express low levels of major histocompatibility complex (MHC)I, lack MHCII and co-stimulatory molecules, it is currently clear that both undifferentiated ESCs and iPSCs, as well as their derivatives, can be rejected after transplantation into an immune-competent recipient (for review see Imberti et al 2015 ). Experiments performed with the use of humanized mice reconstituted with a functional human immune system revealed that even in case of autologous transplantation, iPSCs can induce an immune response leading to cell rejection (Zhao et al 2015 ).…”
