Polarized integrin mediates human keratinocyte adhesion to basal lamina.

Abstract: Epithelial cell interactions with matrices are critical to tissue organization. Indirect immunofluorescence and immunoprecipitations of cell lysates prepared from stratified cultures of human epidermal cells showed that the major integrins expressed by keratinocytes are ad3,4 (also called a644) and a2pl/a3.l1. The adP4 integrin is localized at the surface of basal cells in contact with the basement membrane, whereas a2fil/a3fl1 integrins are absent from the basal surface and are localized only on the lateral s… Show more

“…Such an interaction may contribute to the maintenance of cell surface polarity and to the stability of the epithelial sheet. In the stratified squamous epithelia, the receptor is expressed exclusively by the cells of the basal layer (2,4,12,24). Thus, expression of c~, is rapidly turned off at the onset of cell differentiation and movement to the upper cell layers (7).…”

“…A similar staining pattern was obtained with the anti-cx~ mAb GOH3, and with the B, mAb 346-11A on mouse skin sections (not shown; see also refs. 4,23,24). The COOH-terminal peptide antibodies generated a different pattern.…”

“…For adhesion assays, performed as previously described, 36 96-well plates were coated with 0.1% BSA in calcium-and magnesium-free phosphate-buffered saline (PBS), 10 g/ml collagen IV (Sigma-Aldrich) in 10 mm acetic acid, 10 g/ml laminin-1 (Sigma-Aldrich) in PBS, or 10 g/ml human fibronectin (Sigma-Aldrich) overnight at 4°C. Keratinocytes were plated (30 000 cells per well) in medium in the absence of serum.…”

“…The cytoplasmic HD plaque components, which include HD1/plectin (3) and the bullous pemphigoid antigen 1 (BP230) (4), act as linkers for elements of the cytoskeleton at the cytoplasmic surface of plasma membrane. The trans-membrane constituents of HDs, which include the ␣ 6 ␤ 4 integrin (5,6) and the bullous pemphigoid antigen 2 (BP180) (7), serve as cell receptors connecting the cell interior to extracellular matrix proteins.…”
