Polycomb Group Protein Displacement and Gene Activation through MSK-Dependent H3K27me3S28 Phosphorylation

Abstract: Epigenetic regulation of chromatin structure is essential for the expression of genes determining cellular specification and function. The Polycomb repressive complex 2 (PRC2) di- and trimethylates histone H3 on lysine 27 (H3K27me2/me3) to establish repression of specific genes in embryonic stem cells and during differentiation. How the Polycomb group (PcG) target genes are regulated by environmental cues and signaling pathways is quite unexplored. Here, we show that the mitogen- and stress-activated kinases (… Show more

“…EGF induction has also been shown to induce rapid histone phosphorylation, namely the "nucleosomal response" (55). The phosphorylation of H3S10 and H3S28 via mitogen-and stressactivated kinases results in PcG displacement at a subset of immediate early and stress response genes by impeding Pc chromodomain binding to H3K27me3 (54,56). However, in our study, we do not observe a global displacement of Cbx7 from chromatin upon EGF treatment (e.g., Fig.…”

“…Protein phosphorylation is well recognized as an important mechanism for the regulation of cellular signaling, and a growing body of literature suggests that phosphorylation regulates chromatin dynamics (24,45,54). Here, we report a novel phosphorylation site on mouse Cbx7 that is highly conserved.…”

“…While this manuscript was under review, Gehani et al (30) reported similar findings in that H3 S28 phosphorylation, mediated by MSK1/2, was important for activation of polycomb target genes in response to mitogenic, stress, and differentiation signals. Their conclusions are completely consistent with our findings.…”

“…One mechanism recently described is stress-induced reversal of polycomb repression by phosphorylation of serine 28 of histone H3 in mammals (14). Other candidates for this function are chromatin regulators that genetically act in opposition to polycomb repressors; these are collectively referred to as trithorax group (TrxG) proteins (8).…”
