Polydopamine Decorated Microneedles with Fe‐MSC‐Derived Nanovesicles Encapsulation for Wound Healing

Abstract: Wound dressing with the capacities of antioxidation, antiinflammation, and efficient angiogenesis induction is expected for effectively promoting wound healing. Herein, a novel core‐shell hyaluronic acid (HA) microneedle (MN) patch with ferrum‐mesenchymal stem cell‐derived artificial nanovesicles (Fe‐MSC‐NVs) and polydopamine nanoparticles (PDA NPs) encapsulated in the needle tips is presented for wound healing. Fe‐MSC‐NVs containing multifunctional therapeutic cytokines are encapsulated in the inner HA core o… Show more

“…Human umbilical vein endothelial cells (HUVECs) (CTCC‐0804‐PC, Meisen, Zhejiang, China) and mouse RAW264.7 (ZQ0098, Zhongqiaoxinzhou Biotech, Shanghai, China) were cultured as previously described. 62 , 63 Briefly, HUVECs and RAW264.7 were cultured in an RPMI 1640 medium (#8121486; Thermo Fisher Scientific, Beijing, China) and DMEM:F12 medium, supplemented with FBS (10%), penicillin‐streptomycin (1%), respectively. Cells were maintained in a cell incubator (5% CO 2 , 37°C).…”

“…The drug release reached a plateau after 12 h and totally about 16.9% Rapa was released within 48 h. ) were the most common ROS types. 31,32 Previous studies proved the H 2 O 2 scavenging ability of PDA; 28,33 however, there were less reports on the OH or O 2…”

“…Hydrogen peroxide (H 2 O 2 ), a hydroxyl radical (˙OH), and a superoxide anion (O 2 ˙ − ) were the most common ROS types. 31,32 Previous studies proved the H 2 O 2 scavenging ability of PDA; 28,33 however, there were less reports on the ˙OH or O 2 ˙ − scavenging effect of PDA. 34 Here, we examined the ROS scavenging properties of mPDA and mPDA@Rapa towards H 2 O 2 , ˙OH, and O 2 ˙ − .…”

“…Chronic wounds are those that fail to follow the normal wound healing sequence and do not achieve abnormal healing. The treatment of diabetic wounds remains a major clinical challenge due to their complex wound healing environment, characterized by imbalanced inflammatory responses, the over-expression of reactive oxygen species, hyperglycemia, a lack of angiogenesis, and an extremely high risk of bacterial infection [ 108 , 109 , 110 ]. This wound environment results in chronic and excessive inflammation at the wound site, recurrent infections, and an inability of the dermis and epidermis to respond to regenerative stimuli.…”
