Polymorphisms in Exon 2 of MHC Class II DRB3 Gene of 10 Domestic Goats in Southwest China

Abstract: Polymorphism of the second exon of the caprine leukocyte antigen-DRB3 gene (CLA-DRB3*02) was investigated in this study. The 285 bp PCR product of 258 individuals from 10 domestic goat breeds in Southwest China was digested with restriction endonucleases PstI and HaeIII and then genotyped. Three alleles and 4 restriction digestion profiles were distinguished by digestion of the PCR fragment by PstI, and 8 alleles and 13 genotypes by HaeIII. For HaeIII restriction enzyme sites, the Chi-square (X 2 ) test showed… Show more

“…Non-significant Chi-square value indicated that the two breeds were homogeneous with respect to genotype frequency at this locus. Contrary to our finding, Zhao et al (2011) reported significant differences in genotype frequencies among ten goat breeds of China. These differences might be due to the fact that Sirohi and Barbari breeds have not phylogenetically diversed much whereas ten goat breeds of China under references have evolutionarily diversed much from one other.…”

“…Sirohi and Barbari on amplification of exon 2 of CLA -DRB3 gene (Plate 1a, b). The PCR product of similar bp size has also been reported by Ahmed and Othman (2006) in Egyptian Goat, Baghizadeh et al (2009) in Raeini Cashmere goat, Hernandez (2011) in goats of the central highlands of Veracruz, Zhao et al (2011) in ten domestic goats in southwest china and Singh et al (2012) in Jamunapari breed of goats.…”

“…MHC is well known for containing a large number of genes and genetic diversity in alleles found in most vertebrates. Its genes are divided into three classes; classes I and II genes exhibit most genetic variation (Baghizadeh et al, 2009, Zhao et al, 2011. Heterozygosity of MHC genes plays a key role in immune recognition of pathogens and parasites hence improved resistance because of diverse antigens that will be presented to T-cells and by generating a diverse collection of T cells Genetic polymorphisms of class II genes occur predominantly in the first domain (exon 2) that encodes the peptide-binding sites (PBS).…”
