“…Forty DNA samples were randomly selected from each cattle breed, to construct for DNA pools of four breeds [ 19 ]. PCR reactions were carried out using a Touchdown PCR System Thermal Cycler Dice (TaKaRa, Dalian, China) and a PCR protocol which was described in a previous study [ 20 ]. DNA pool sequencing for the PCR productions using a Sanger sequencing strategy was applied to screen the genetic variations.…”

“…The PCR reactions were carried out in a 25 μL volume which contained 1 μL cattle genomic DNA (50 ng/μL), 0.5 μL each primer (10 μmol/L), 12.5 μL 2 × Taq PCR MasterMix (Taq DNA polymerase, Mg 2+ , dNTPs, et al), and ddH 2 O. The PCR tests were carried out with the PCR System Thermal Cycler Dice (TaKaRa, Dalian, China), with the following amplification protocol: 94 °C for 5 min; followed by 35 cycles of 94 °C for 30 s, annealing at selected temperatures ( Table 1 ) for 30 s, and extension at 72 °C for a 30 s; and a final extension at 72 °C for 10 min [ 20 ].…”

“…Genetic characterisation of a number of Chinese cattle breeds, which historically exhibit small body size and low intramuscular fat content compared to other meat production breeds, shows that three polymorphisms in Flii are associated with increased body mass, height and length, as well as chest girth. Moreover, these polymorphisms; CT (rs41910826), TT (rs444484913), and CA (rs522737248) were associated with increased PPARγ in adult adipose as well as increased Flii in fetal muscle (Liu et al, 2016).…”

“…Flii has been shown to act as a repressor of the transcriptional activity of the Peroxisome Proliferator-Activated Receptor γ (PPARγ) and its overexpression suppresses adipogenesis (Choi et al, 2015). Flii is expressed more highly in adult bovine adipose tissue compared to fetal bovine adipose tissue (Zhou et al, 2014), with Flii expression increasing in differentiated adipocytes compared with preadipocytes (Liu et al, 2016). In the absence of ligand, the LXXLL motif within the LRR domain of Flii binds directly to the DNA binding domain of the receptor to prevent PPARγ receptor occupancy at the promotor of target genes as well as blocking the interaction between PPARγ and retinoid X receptor α (RXRα) (Choi et al, 2015).…”

“…Finally, the CNV results were summarized and displayed by Prism software (version 5.0.0). In the analysis of gene expression profiles, internal reference Emerin gene (EMD) and lipoprotein receptor-related protein 10 gene (LRP10) were used as reference internal genes for the expression level identifications following the study of Liu et al [47] and Saremi et al [48], respectively. The gene expression levels were also The amplification efficiencies of primers were tested by genomic DNA with PCR products concentration gradients that were 500, 100, 20, 4 and 0.8 ng.…”
