Polypeptide sequence of the chlorophyll a/b/c-binding protein of the prasinophycean alga Mantoniella squamata

Abstract: The primary structure of the Chla/b/c-binding protein from Mantoniella squamata is determined. This is the first report that protein sequencing reveals one modified amino acid resulting in a LHCP-specific TFA-cleavage site. The comparison of the sequence of Mantoniella with other Chla/b-and Chla/c-binding proteins shows that the modified amino acid is located in a region which is highly conserved in all these proteins. The alignment also reveals that the LHCP of Mantoniella is related to the Chla/b-binding pro… Show more

“…The alignment of amino acid sequences reveals a phylogenetic relationship of this apoprotein to Chl ah-binding polypeptides (29). One has to remember that in accordance with recent evidence, P. marinus and P. didemni contain a Chl c-like pigment together with (divinyl) Chls a and b (7,15).…”

“…The SCP MLHC and DM MLHC showed the typical LHC apoproteins of M. squamata with MWs of 23.5 and 25 kDa (Fig. 2D, Schmitt et al 1994). The SCP MLHC was purer and no additional polypeptide bands were found in the SDS-PAGE gels.…”

“…Whereas most of these algae do not contain Chl b , some Prasinophycean algae, such as Montoniella squamata , accumulate a Chl c , 3,8-divinyl-Pchlide, along with Chls a and b (Schmitt et al. 1994 ; Green and Durnford 1996 ). The red alga that contributed its plastid in the secondary endosymbiotic event apparently lacked Chl c .…”

“…The Chl-binding proteins in these organisms are evolutionarily related to those in green algae and plants (Schmitt et al. 1994 ; Green and Durnford 1996 ). The relatively simple LHCs are well-defined structures and consequently are excellent systems to search for basic mechanisms.…”
