Poor Hypo-Osmotic Swelling Test Results from Cryopreserved Sperm Despite Preservation of Sperm Motility

Abstract: Cryopreservationithawing of sperm leads to decreased motile density. Most donor programs select for cryopreservation a male with a high initial motile density and if the post-thaw semen has a motile density over 10 x 106/ml, the specimen is thought to represent a fertile specimen. Some recent data suggest that males with normal motile densities but subnormal hypo-osmotic swelling (HOS) tests may be infertile. A study was thus performed to bee if males with an adequate motile density after cryopreservation may … Show more

“…However, computer assisted motility of bovine sperm, immediately after thawing is not correlated with fertility 23. Similarly Lapointe et al24 suggested the beneficial effect of 0.1 mM MnCl 2 for the maintainace of sperm motility without detrimental effects on mucus penetration and fertilizing ability and hypothesized that Mn ++ would have an effect on sperm cyclase activity that lead to increased Ca ++ concentration and motility.…”

Liquefied semen (50 mL) was mixed with 500 mL of different hypoosmotic solutions for 5 minutes at room temperature.…”

“…Though there have been divergent degrees of correlation reported between the HOS test and the different sperm evaluation parameters, the test has been recognized as the predictor of human sperm fertility (4)(5)(6)(7). Though it is a very simple and sensitive test, it has never been routinely practiced.…”

“…No entanto, com base nos resultados das sondas fluorescentes, esperava-se que esses gametas apresentassem sinais de hiperativação, devido à ocorrência de danos na membrana plasmática causados pela congelação (CHECK & CHECK, 1991), resultando no aumento da quantidade de espermatozoides capacitados (MAXWELL & WATSON, 1996).…”
