Population-specific coding variant underlies genome-wide association with adiponectin level

Abstract: Adiponectin is a protein hormone that can affect major metabolic processes including glucose regulation and fat metabolism. Our previous genome-wide association (GWA) study of circulating plasma adiponectin levels in Filipino women from the Cebu Longitudinal Health and Nutrition Survey (CLHNS) detected a 100 kb two-SNP haplotype at KNG1-ADIPOQ associated with reduced adiponectin (frequency = 0.050, P = 1.8 × 10(-25)). Subsequent genotyping of CLHNS young adult offspring detected an uncommon variant [minor alle… Show more

“…Top SNPs in these studies were consistent with top SNPs in our study, although it should be noted that two of the studies were previously performed in the InCHIANTI population, which was also included in this analysis. Ultimately, haplotype analyses with denser SNP panels, direct sequencing and functional analyses will be required to localize the causal variants in this region (38). …”

“…None of the receptor SNPs was associated with serum adiponectin in this study [27]. Furthermore, rare population-specific mutations have been shown to account for ~17% of the variance in serum adiponectin concentrations ( Table 1) [28][29].…”

“…However, this would be rather an indirect/downstream effect on serum leptin levels than an effect of polymorphisms directly acting on LEP gene transcription and protein expression, and the hypothesis is not yet proven. In contrast, in a recent but small study, [23], [28], rev. …”
