Pore formation: An ancient yet complex form of attack

Abstract: Bacteria, as well as higher organisms such as sea anemones or earthworms, have developed sophisticated virulence factors such as the pore-forming toxins (PFTs) to mount their attack against the host. One of the most fascinating aspects of PFTs is that they can adopt a water-soluble form at the beginning of their lifetime and become an integral transmembrane protein in the membrane of the target cells. There is a growing understanding of the sequence of events and the various conformational changes undergone by… Show more

“…The majority of bacterial PFTs are ␤-PFTs, which also form the most-studied group (227). The large-pore-forming cholesterol-dependent cytolysins (CDCs), produced by Gram-positive and some Gram-negative bacteria, are a ␤-PFT subclass (4,227,228).…”

“…P athogenic bacteria secrete various virulence factors to attack host cells. The pore-forming toxins (PFTs) are among the most sophisticated virulence factors, and are expressed as watersoluble monomeric proteins that assemble on the membranes of the target cells to form bilayer-spanning pores (1). With the appearance of the pore on the membrane, the cells are killed through leakage.…”

“…Although VCC's involvement as a virulence factor in the human cholera pandemic remains unclear, this toxin belongs to a much larger class of pore-forming toxins (PFTs) secreted by a wide variety of human pathogens (5). PFTs are released by bacteria as water-soluble species and can attack cells at a distance, especially if they enter into the bloodstream.…”
