Post-Mortem Tissue Biopsies Obtained at Minimally Invasive Autopsy: An RNA-Quality Analysis

Abstract: IntroductionBereaved relatives often refuse to give consent for post-mortem investigation of deceased cancer patients, mainly because of the mutilation due to conventional autopsy (CA). Minimally invasive autopsy (MIA) may be a more acceptable alternative and, if implemented in clinical practice, creates an opportunity to more often obtain post-mortem tissue samples of (recurred) primary tumors and metastases for molecular research. As a measure for tissue quality for molecular studies, we hereby present a fea… Show more

“…In this study, we could detect a control RNA target in more than 90% of the study samples suggesting a reasonable RNA detection sensitivity and integrity in our FFPE material. Other studies have established that FFPE tissue material is usable for reverse-transcriptase PCR analysis of virus and messenger RNA, although the analytical sensitivity is reduced when compared to the use of matched frozen tissue material [12].…”

“…Schoor et al [40] and van der Linden et al [47] stated that while all efforts should be made to obtain high quality RNA samples that reflect the natural state most reliably, partially degraded samples may still lead to a reasonable expression profile. Therefore, due to all of these aspects, we believe that RNA integrity did not have a high impact in the expression profiling and analysis of the present work.…”
