Potential Role of Nanoparticles in Treating the Accumulation of Amyloid-Beta Peptide in Alzheimer’s Patients

Abstract: The disorder of Alzheimer’s is marked by progressive pathophysiological neurodegeneration. The amino acid peptides in the amyloid plaques found in the brains of people with Alzheimer’s disease (AD) are known as amyloid-beta (Aβ). Current treatments are not curative, and the effects associated with AD are reduced. Improving treatment results involved the targeting of drugs at optimum therapeutic concentration. Nanotechnology is seen as an unconventional, modern technology that plays a key role in the treatment … Show more

“…In another study, β-amyloid fibril formation was observed and it was found that TiO 2 -NPs could promote β-amyloid fibrillation by shortening the nucleation process, which is the key rate-determining step of fibrillation [ 57 ]. Thus, the potential role of nanoparticles in treating the accumulation of amyloid-beta peptide in Alzheimer’s patients was reviewed. It was noticed that the nanoparticles from three different categories, i.e., polymer, lipid, and gold nanoparticles, could prevent the accumulation of β-amyloid during the efficient delivery of the drug to the cells to treat Alzheimer’s disease [ 58 ].…”

“…However, these small molecules and natural products are limited by fast hepatic metabolism and poor BBB permeation [219]. Nanotechnology offers the advantage over the conventional therapeutic approaches by stabilizing drugs to cross the BBB and exhibit a superior pharmacotherapeutic effect [220]. However, this is not always the case.…”

“…The delicate balance between nerve connections, spinal cord, and the control center, the brain, is easily perturbed by exogenous insults and endogenous degenerative conditions. Debilitating disorders of the central nervous system (CNS) include Alzheimer's disease (AD) [17] and variants of dementia such as Lewy body dementia (LBD), tumors such as neuroblastoma and glioblastoma [18], as well as dyskinesia-related conditions such as Parkinson's disease (PD), Huntington's disease (HD), and amyotrophic lateral sclerosis (ALS) [19]. Alzheimer's disease is a type of neurodegenerative disease that is described by the formation of plaques with tangled neurofibers composed of amyloid and tau proteins.…”
