“…Similar concern had been reported recently regarding the poor correlation of potency results in cord blood stem cells obtained from attached segments with those from the corresponding cord blood units . This stands in contrast to many authors who assume that post‐thaw flow cytometric analyses reflect the composition of cells infused and can be used to draw conclusions on the outcome in terms of time to engraftment . This seems to be justified in some, but not in all cases.…”

“…The use of post‐thaw flow cytometric potency testing is divisive but performed in many centres. While it is well accepted that the composition of the cryopreservation solution plays an important role in post‐thaw cell recovery , the influence of temperature during the 30 min staining period on cell survival is not generally known and reported post‐thaw recovery results are rather controversial . We were able to show highly significant differences in viable cell recovery when thawed cells were stained at 4°C vs. RT.…”

“…Regarding engraftment, we found that higher postthaw CD34+ cell counts were associated with faster engraftment of ANCs, PLTs, and RBCs. PLT engraftment was the most sensitive parameter, as already described by others . For completeness of cellular engraftment data, we also assessed RBC engraftment, and found that it depended on CD34+ count infused.…”
