Prenatal Curcumin Administration Reverses Behavioral and Neurochemical Effects and Decreases iNOS and COX-2 Expressions in Ischemic Rat Pups

Abstract: The objectives were to evaluate alterations in ischemic rat pups, from dams administered with curcumin (25 and 50 mg/kg). Ten-day-old male pups were subjected or not (SO) to brain ischemia and reperfusion, for 1, 7, and 14 days (this last group was submitted to behavioral evaluation). After that, pups were euthanized for determinations of striatal DA and DOPAC in SO, ischemic from untreated (ICUD) or curcumin treated (ICTD) dams, as well as hippocampal immunohistochemistry assays for iNOS and COX-2 and cresyl … Show more

“…Nissl bodies (staining with cresyl violet) are an indicator of neuronal integrity and may be lost during neuronal damage, which aids in determining the number of viable neurons (Telles et al. 2014 ). Marked neuronal changes were observed in HI insulted rats with increased pyknotic neurons and less visible Nissl bodies (increased neuronal death).…”

“…Our results are well supported by Tan and his coworkers (2011), also pointed out that MF can effectively reduce the cerebral infarct size in a cerebral ischemic rat model by improving the antioxidant status and thus preserving the integrity of BBB. The degree of ischemic neuronal injury was determined by checking the number of normal motor neurons using cresyl violet staining procedure based on the presence of nissl bodies (Telles et al, 2014). Enormous changes in neuronal morphology were noticed in a hypoxic-ischemic group with pyknotic neurons as well as few visible nissl bodies, thus indicating the neurotoxicity in HI insult condition with reduced viable neuronal cells.…”
