Preparation and characterization of amnion hydrogel and its synergistic effect with adipose derived stem cells towards IL1β activated chondrocytes

Abstract: Inflammation leads to chondrocyte senescence and cartilage degeneration, resulting in osteoarthritis (OA). Adipose‐derived stem cells (ADSCs) exert paracrine effects protecting chondrocytes from degenerative changes. However, the lack of optimum delivery systems for ADSCs limits its use in the clinic. The use of extracellular matrix based injectable hydrogels has gained increased attention due to their unique properties. In the present study, we developed hydrogels from amnion tissue as a delivery system for A… Show more

“…An increase in PRG4 upon IL-1β–induced inflammation was observed and may indicate a negative feedback regulation of inflammation. Interestingly, in addition to having a similar antiinflammatory effect compared to ADSCs [reducing inflammatory effects ADAMTS5 ( 49 , 50 )], SASC was able to reverse the effects of inflammation on NO, SOX9, and PRG4 ( Fig. 4 A – D ).…”

“…Lastly, with advancements towards 3-dimensional cell printing [55,[64][65][66], it would be interesting to evaluate the effect of printing the SVF-like extracellular matrix in a hydrogel scaffold for osteochondral defects, or incorporating SVF into an injectable hydrogel for enhanced retention and viability in the local environment as has been demonstrated in previous studies using adipose-derived stem cells [67].…”
