Preparation and partial characterization of cell-free protein-synthesizing extracts from Tetrahymena pyriformis

Abstract: 1. We have examined methods necessary for preparing post-mitochondrial supernatants from Tetrahymena pyriformis strain HSM that are capable of efficient cell-free protein synthesis. 2. The requirements for optimum synthesis in these extracts are described. 3. Data relating to the kinetics of protein synthesis and the initiation capacity of these supernatants are presented.

“…Exponential-phase cultures of Tetrahymena were harvested and used for preparing the postmitochondrial extracts as described previously (David & Smith, 1981, in the section on 'Final procedures'), except that the preparation buffer used comprised 25 mM-KCl, 7 mM-magnesium acetate, 1 mM-dithiothreitol, 0.lmM-EDTA, 1.5mM-EGTA and 10% (v/v) glycerol made up in 20mM-triethanolamine/ HCl buffer, pH 7.4.…”

“…(1) Theoretically, the most satisfying solution is a homologous message-dependent translation system. A Tetrahymena system for translation in vitro has been described (David & Smith, 1981), but it is dependent on endogenous mRNA and has a poor initiation capacity.…”

“…It appears that hypotrichs use the unusual codons rarely, Tetrahymena often, and Paramecium prefers them over the normal ones. These observations account for the poor behavior of Tetrahymena messengers in heterologous systems of in vitro protein synthesis (5) and the almost complete lack of synthesis observed when Paramecium messengers are used ( 1 9).…”
