Preparation, characterization and anticancer activities of resveratrol loaded redox-sensitive F127-SS-TOC micelles

Abstract: Resveratrol (RES) loaded redox-sensitive F127-SS-TOC micelles (F127-SS-TOC/RES) hold the potential in improving the treatment of breast cancer, which compared to free RES or non-redox-sensitive micelles (F127-TOC/RES).

“…It was different from some reports, this phenomenon might be attribute to sustained drug release rate. 38 The speculation was conrmed in the FAP/CUR micelles group and the MIX/CUR micelles group. Because they were formed by FAP segments, so they can be hydrolyzed in a weakly acid environment to increase the release rate of CUR from micelles.…”

“…DCFH-DA could pass the membrane into the cells and be cleaved by esterase inside the cells to form 2′,7′-dichlorodihydrofluorescein (DCFH), which was further oxidized by ROS to form dichlorofluorescein (DCF) which has green fluorescence. 38 In short words, Hepa1-6 and SMMC-7721 cells (8000 per well) were placed in 96-well plate in 100 μL DMEM medium and incubated overnight. The cells were treated with free CUR, FBP/CUR, FAP/CUR and MIX/CUR micelles (equivalent concentration of 50 μM CUR for SMMC-7721 cells and 25 μM CUR for Hepa1-6 cells), respectively.…”

“…Once the Pluronic F127-SS-TOC polymeric carrier loaded with the encapsulated compound is taken up by the cancer cell via endocytosis, the S─S bonds between the polymers will be cleaved in the presence of high intracellular concentration of GSH (2-10 mM), thereby the polymeric carrier destabilizes, leading to triggered release of the encapsulated compound into the cytosol nanocarriers, which can easily be broken (reduced) by reducing agents, such as GSH and dithiothreitol (DTT), leading to carrier deformation and core release. Liu et al synthesized redox-sensitive micelles with Pluronic F127-SS-α-tocopherol (TOC) polymer for the delivery of Res to breast cancer cells (Liu et al, 2017). The micelles (F127-SS-TOC/Res) were stable under ambient conditions for up to 7 days.…”
