The purpose of this study was to develop and characterize hydroxypropyl-βcyclodextrin (HPβCD) hybrid nanogels for solubility enhancement of lipophilic drug, dexibuprofen. HPβCD hybrid nanogels were designed by chemical crosslinking of hydroxypropyl-β-cyclodextrin with 2-acrylamido-2-methyl propane sulfonic acid (AMPS) via free radical polymerization. Loading efficiency, solubilization, zetasizer, FESEM Fourier transform infrared (FTIR), differential scanning calorimetry (DSC), thermal gravimetric analysis (TGA), powder X-ray diffraction (PXRD), and in vitro drug release analysis were performed for characterization of hybrid nanogels. Efficient solubilization of dexibuprofen has been observed by HPβCD hybrid nanogels. FTIR, TGA, and DSC studies confirmed the formation of new hybrid nanogels and complexation of dexibuprofen with nanogels. XRD analysis revealed loss of dexibuprofen crystallinity in hybrid nanogels. Highly porous and amorphous nanogels showed a significant dexibuprofen release in aqueous medium. In toxicity studies, no significant changes in behavioral, physiological, biochemical, or histopathological parameters of animals endorsed that developed formulations are nontoxic and biocompatible. Designing Aptamer-Gold Nanoparticle-Loaded pH-Sensitive Liposomes Encapsulate Morin for Treating Cancer

This study proposes the synthesis of a type of anticancer nanoparticle, aptamers and Au nanoparticle (Apt-Au)modified Morin pH-sensitive liposome (MSL), which exhibits targeting properties. Tumors are difficult to cure because their microenvironment varies from that of normal tissue; its pH is lower than that of normal tissue, which generally impedes the effectiveness of drugs. Thus, pH-responsive drugs have attracted extensive attention. Gold nanoparticles (AuNPs) show potential as drug carriers because of their small size, good biocompatibility, easy surface modification, and strong cell penetration. Apt-Au@MSL exhibits excellent monodispersity and tumor-targeting properties and can be released in partly acidic environment via dialysis. We screened our model cancer cell by MTT assay and found that SGC-7901 cells can effectively suppress proliferation. In vivo results demonstrate that the administration of Apt-Au@MSL could inhibit tumor growth in xenograft mouse models. H&E staining and TUNEL assay further confirmed that Apt-Au@MSL can promote tumor apoptosis. Apt-Au@MSL may induce apoptosis by triggering overproduction of reactive oxygen species (ROS) and regulating multiple signal crosstalk. Both blood biochemistry tests and H&E staining suggested that these materials exhibit negligible acute toxicity and good biocompatibility in vivo. With its powerful function, Apt-Au@MSL can be used as a target-based anticancer material for future clinical cancer treatment.

Herbal medicines (HMs) are gaining increasing popularity and recognition worldwide due to their eco‐friendliness and efficacy. With their multi‐compounds, multi‐targets, and multi‐pathways characteristics, HMs have been used in treatment of various diseases. However, the clinical applications of preparations containing HMs have been limited due to their inherent physicochemical properties, including low water solubility, poor stability, and unsatisfactory bioavailability of bioactive compounds. Supramolecular macrocyclic hosts, like cyclodextrins, calixarenes, cucurbiturils, and pillararenes, are important objects of researches in supramolecular chemistry. These hosts have been utilized to encapsulate the ingredients, improve the solubility of poorly water‐soluble components, enhance the stability of the tested compounds, increase the bioavailability of bioactive compounds, and ensure the safety of HMs. Herein, we provide a brief introduction to the theories of supramolecular chemistry and summarize the extensive applications of supramolecular macrocyclic hosts in the field of HMs. These applications encompass the screening of bioactive compounds in HMs and the enhancement of druggability for HMs. We hope this review can provide a strategy for dealing with the challenges of HMs, thereby enabling their better applications and development.
