Preparation of uniform-sized GeXIVA[1,2]-loaded PLGA microspheres as long-effective release system with high encapsulation efficiency

Abstract: The purpose of this study was to prepare GeXIVA[1,2] PLGA microspheres by W/O/W re-emulsification-solvent evaporation technology and to develop sustained-release formulations to meet the clinical treatment needs of chronic neuropathic pain. Through prescription optimization, the uniformity of particle size and the encapsulation efficiency is improved, so as to achieve the quality standard of the microspheres. The mechanism of trehalose improving the stability of GeXIVA[1,2] was studied and verified by molecula… Show more

“…On the contrary, Li et al. indicate that when the PVA concentration increases, the particle size decreases, the specific surface area increases and the particles become more uniform, that is, low size distributions are obtained (Li et al., 2022 ).…”

“…Moreover, drug development for clinical use requires working out a stable medicinal formulation. In the case of conotoxins, lyophilization or encapsulation in microspheres could diminish their intrinsic instability and provide a sustained release of the drug to meet the clinical treatment needs of chronic pain [ 207 , 233 ]. In addition, new small-molecules [ 182 , 234 ] and peptide [ 235 ] inhibitors of α9-containing nAChRs are emerging, differing in chemical structure from known analogs.…”

“…In the preparation process, the addition of NaCl in the outer aqueous phases can increase the osmotic pressure to inhibit the mass exchange of the inner and outer phases of the microemulsion, which in turn affects the structure and permeability of microspheres. 32,33 Generally, higher osmotic pressure resulted in a denser internal structure. Consistently, 5% NaCl significantly increased the DL and EE of microspheres (Fig.…”
