Pretreatment of Cell Suspensions as a Method to Increase the Protoplast Yield of Haplopappus gracilis

Abstract: A low yield of free protoplasts was obtained when fast‐growing cell suspensions of Haplopappus gracilis were treated with Driselase, an active mixture of cellulase and pectinase. If the cells in the suspension culture were pretreated by increased levels of auxin, reduced sugar concentration, addition of sulphur‐containing amino acids, or by the reducing agent mercaptoethanol, protoplasts were released from a higher proportion of the cells. These pretreatments did not adversely effect division of the protoplast… Show more

“…The effective osmotic concentration will depend on the endogenous cell osmotic pressures at the time of isolation and these are markedly influenced by environmental growth conditions (Shepard and Totten, 1977;Wallin et al, 1977) and age (Kao and Michayluk, 1980) of donor tissues.…”

“…1970), Acer pseudoplatanus (Bright and Northcote 1974) and Nicotiana tabacum (Uchimiya and Murashige 1976) just to mention some of the more frequently used dicots. In others, like Haplopappus gracilis (Wallin et al 1977) or Rosa 'Paul's Scarlet' (Pearce and Cocking 1973), protoplasts were isolated but without regeneration to the callus stage.…”
