As addiction is increasingly formulated as a developmental disorder, identifying how early developmental exposures influence later responses to drugs of abuse is important to our understanding of substance abuse neurobiology. We have previously identified behavioral changes in adult mice following gestational exposure to cocaine that differ when assessed with methods employing contingent and non-contingent drug administration. We sought to clarify this distinction using a Pavlovian behavioral measure, conditioned place-preference. Adult mice exposed to cocaine in utero (40 or 20 mg/kg/day), vehicle and pair-fed controls were place-conditioned to either cocaine (5 mg/kg or 20 mg/kg, i.p.) or saline injections. The development of conditioned place-preference to cocaine was impaired in mice exposed to cocaine in utero, and was abolished by fetal malnutrition. A context-specific place-aversion to vehicle but not cocaine injection was observed in prenatally cocaine-exposed mice. Locomotor behavior did not differ among prenatal treatment groups. We conclude that early developmental exposure to cocaine may diminish the subsequent rewarding effects of cocaine in adulthood measured with classical conditioning techniques, and that this is not due to changes in locomotor behavior. Sensitivity to acute stress is also altered by prenatal cocaine exposure, consistent with earlier findings in this model.

Infant rats emerge from the maternal nest at Postnatal Day 17-18 to have their first critical environmental experiences; they may be particularly sensitive to experiences or experimental interventions that can affect their adult capacity. The authors address open questions on 2 components of normative environmental exploration, locomotor activity and response to anxiety-provoking locations, in Postnatal Day 18 infant and Postnatal Day 60 adult rats. The authors compare diurnal patterns of locomotor activity, wheel running, novel and familiar open-field activity, and 2 measures of anxiety. Infants have an equivalent capacity to adults for locomotor activity and wheel running and a fundamentally adult-like diurnal rhythm, except that they do not anticipate light-dark transitions, are more perturbable at their most somnolent, and are more or less active during specific limited phases than adults. Infants initially have a lower rate of locomotor activity in novel environments and have a greater willingness to be active in anxiety-provoking locations. Such differences may allow enhanced gathering of environmental information by the infant and are important to consider in the design of experiments using infants. Keywordsinfant; adult; exploration; activity; anxiety In humans, late childhood and adolescence is the period when drug sampling and experimentation first begins (Brown & Tapert 2004;Johnston, O'Malley, Bachman, & Schulenberg, 2005;Spear, 2000;Turner, Mermelstein, & Flay, 2004), when clinical mental health disorders requiring pharmacologic intervention, such as attention-deficit/hyperactivity disorder, emerge (Andersen, 2003;Andersen, Arvanitogiannis, Pliakas, LeBlanc, & Carlezon, 2002), and when the central nervous system (CNS) is readily shaped by many stimuli, often with long-lasting consequences. Ultimately, studies using immature animals may inform us about human disorders with early life origins (e.g., hyperactivity) and may inform the beneficial use of pharmacological treatments. Further, these studies may provide insight into the effects of preadolescent sampling of drugs of abuse (Dahl & Spear, 2004).Prior to achieving sexual maturity between Postnatal Days (P) 55-60, immature rats are either infants (P1-P28) or adolescents (P28-P55; Ojeda & Urbanski, 1988;Spear, 2000). Infant rats are vigilantly kept in the protective maternal nest by their mothers until around P17-P18 (Barnett, 1958;Galef & Clark, 1972; Pereira & Morrell, 2007;Small, 1899), when they are allowed out into their larger environment. At this point, they have full motor and sensory capacity (Moorcroft, Lyttle, & Campbell, 1971). The rate of acquisition of drug self-administration may serve as a predictor of later drug-taking behavior, possibly influencing the vulnerability to use drugs. The present study examined the effects of perinatal (gestation/lactation) lead exposure on adult rates of acquisition of intravenous cocaine self-administration using an automated procedure that included both Pavlovian and operant components. For Experiment 1, female rats were gavaged daily with 0 or 16 mg lead for 30 days prior to breeding with nonexposed males. Metal administration continued through pregnancy and lactation and was discontinued at weaning (postnatal day (PND) 21). Animals born to control or lead-exposed dams subsequently were tested daily as adults in a preparation where sessions included an initial 3-h autoshaping period followed by a 3-h self-administration period where 0.20 mg/kg cocaine was delivered contingently. During autoshaping, intravenous cocaine infusions were paired with the extension and retraction of a lever, while infusions occurred during selfadministration only when a lever press was executed (FR-1). The criterion for acquisition was a 2-day period during which a mean of 50 infusions/session occurred during self-administration. Animals were given 35 days to reach criterion. In Experiment 1, accelerated rates of acquisition of cocaine self-administration were evident for lead-exposed animals relative to controls. Overall, the number of selfadministered cocaine infusions per session was significantly higher for lead-exposed rats as compared to control rats. Experiment 2 replicated Experiment 1 except that a higher dose of cocaine (0.80 mg/kg) was employed as the reinforcer, and 30 infusions/session was the set criterion. At the higher cocaine dose (Experiment 2), acquisition rates for control and lead-exposed animals were not markedly different, and significantly different infusion rates were not observed.
