ProAKAP4 Concentration Is Related to Sperm Motility and Motile Sperm Subpopulations in Frozen–Thawed Horse Semen

Abstract: ProAKAP4 is the precursor of AKAP4 (A-kinase Anchor protein 4), the main structural protein of the fibrous sheath of sperm. The amount of proAKAP4 reflects the ability of spermatozoa to maintain the flagellum activity and functionality up to the site of fertilization and is positively correlated with progressive motility in several mammalian species. “…For example, in horses, the first reported values for frozen–thawed semen were between 3 and 12 ng/mL (isolated from around 7.5 × 10 6 spermatozoa), while 2 years later, the same team in another study obtained a mean concentration 60.9 ± 2.1 ng/10 million spermatozoa (Blommaert et al., 2019, 2021). Another study of frozen–thawed horse semen classified values of 5.06–35.61 ng/10 million spermatozoa as ‘low’ and values of 39.92–82.23 ng/10 million spermatozoa as ‘high’ (Dordas‐Perpinyà, Yánez‐Ortiz, et al., 2022).
