Probiotics as an Adjuvant for Management of Gastrointestinal Cancers through their Anti-inflammatory Effects: A Mechanistic Review

Abstract: The immune system's role in maintaining the health of the gastrointestinal (GI) system is like a double-edged sword. Simultaneously, it could reduce the risk of pathogen invasion by the inflammatory response. But, if regulated improperly, it could also propagate oncogenic signaling that transfers a normal cell into the malignant counterpart. Thus far, several mechanisms have been proposed, such as the immune system could disturb the GI homeostasis and increase the survival and proliferative capacity of cells l… Show more

“…Therefore, a better understanding of PCSK9's role in the cancerous pathways is extremely important ( 7 ). Since this marker has a significant role in different parts of body, it has been used as a pharmacological target in the past few years ( 8 - 10 ).…”

“…GIT probiotics naturally regulate the growth of other microorganisms, intestinal permeability, digestion, and metabolism. They are also connected to human physiology through crosstalk with different body sites and the immune system, driving changes in the response to drug therapies as well as the prevention, onset, and progression of several diseases including cancer ( Javid et al, 2023 ; Nami et al, 2023 ) and non-cancer conditions ( Un-Nisa et al, 2023 ; Xia et al, 2023 ).…”

“…In comparison with biomaterial-based drug carriers, probiotics are characterized by sustainability, scalability, relatively low cost, inherent biotargeting ability with possible spacio-temporal control over drug release ( Xie et al, 2021 ). Moreover, a properly selected probiotic may act as therapeutic adjuvant to improve the efficacy and safety of the probiotic-cargo combination and restore homeostasis within a perturbed microbial community ( Javid et al, 2023 ).…”

“…[8][9][10] Metastasis is a multistage process that disseminates cancer cells from primary tumors and establishes secondary lesions in distant parts of the body. 11,12 The metastatic mechanism can be formalized in a variety of discrete phases. This process involves five steps: local invasion of tumor cells into the surrounding matrix (Step 1), intravasation of tumor cells into the circulatory system (Step 2), the systemic transmission of tumor cells (Step 3), extravasation of tumor cells into the parenchyma of distant tissue sites (Step 4), colonization of distant organs, and macroscopic tumor establishment (Step 5) (Figure 1).…”

“…Cancer metastasis is the leading cause of cancer mortality and morbidity, accounting for about 90% of deaths from cancer 8–10 . The metastatic mechanism can be formalized in a variety of discrete phases.…”
