Processable Cyclic Peptide Nanotubes with Tunable Interiors

Abstract: A facile route to generate cyclic peptide nanotubes with tunable interiors is presented. By incorporating 3-amino-2-methylbenzoic acid in the D,L-alternating primary sequence of a cyclic peptide, a functional group can be presented in the interior of the nanotubes without compromising the formation of high aspect ratio nanotubes. The new design of such a cyclic peptide also enables one to modulate the nanotube growth process to be compatible with the polymer processing window without compromising the formation… Show more

“…Given the pharmaceutical (44) and nanotechnological (45,46) applications for cyclic peptides, it was of interest to further test the range of possible CP, which could be produced by PCY1. Two presegetalin A1 [14,32] variants representing insertions of A and AAA in the incipient CP sequence were tested ( Table 1).…”

“…20 As DLCPs are synthesized via common solid-state peptide synthesis methods 24 , one has the freedom to explore limitless combinations of amino acids and postsynthetic modifications. 25 This versatility has been leveraged to study DLCPs in a variety of applications including structural antibiotics, 26 ion channels, 27 selective ion transporters, 28 and reinforcing agents. 29 Additional research has focused on the fundamental properties of cyclic peptides including the thermodynamics of assembly and the breadth of structural flexibility.…”

“…5 Since then formation of nanotubes and other structures such as nanorods, nanofibers, spherical structures from various cyclic peptides 6 and their potential application had been reported thereof. 7 In recent example by Ghadiri et al self-assembled nanotubes from cyclic peptides could block entry of Hepatitis C virus through cell membrane. 8 In another report transport of antitumor drug through cyclic peptide nanotube was shown.…”

“…6 Growing interest for transportation through cyclic peptide nanotubes had resulted in modifications of them with unnatural amino acids such as β-amino acids, alternating α-and β-amino acids, alternating α-and γ-amino acids, δ-amino acids and carbohydrates, 10 to have specific binding site at the interior of the nanotubes. 7 Although having significant utilities, limitation of cyclic peptides arises from difficulty in their synthesis, low yield and requirement of expensive coupling reagents. 1 Our group is concerned with the synthesis and design of various peptide based molecules for creation of nanostructured materials for applications such as drug delivery, multivalent receptors etc.…”
