“…hASCs were isolated as previously described (40) and kindly provided by Dr. Julie Fradette (Université Laval, LOEX, Quebec City, Quebec). More specifically, as approved by the ethics committee, these ASCs were isolated from subcutaneous fat lipoaspirated for cosmetic purposes from three women aged 38, 40, and 46 (identified as LA38, LA40, and LA46, and in this study as donor 1, 2, and 3, respectively) with a mean body mass index of 22.4 Ϯ 2.8 (SD).…”

“…Although the differentiated 3T3-L1 cells never develop into fully mature adipose cells and are maintained in culture only for a relatively short period after differentiation, mature adipose cells can be cultured only for a few days after isolation, which limits the application of techniques such as small interfering RNA. New strategies involving stromal cells derived from adipose tissue have shown a potential to develop into mature adipose cells that could be kept in culture without losing the adipose phenotype (28). This promises to be an invaluable tool for answering the question how differentiation and maturation of adipocytes affects molecular machinery involved in GLUT4 exocytosis.…”

“…For comparative purposes, reconstructed connective tissue sheets devoid of adipocytes were also produced by using the standard medium throughout the culture period. 32 Tissue reconstruction proceeded as a multistep protocol based on adaptations of previously published methods from our group. 32,33,35 The adipose substitutes were produced according to two main models based on initial cell seeding into six-well plates for series 1 and 2 (9.6 cm 2 ), while series 3 consisted of a model produced from initially larger culture vessels (57-87 cm 2 ).…”

“…The resulting connective cell sheets can then be superposed to produce thicker connective tissues. 32,33 If an adipogenic induction step is performed in parallel to matrix stimulation, then the resulting tissue sheets feature functional adipocytes filled with lipids. 32,34 These reconstructed tissues do not contain synthetic or exogenous biomaterials, and their production can be customized for shape and thickness.…”
