Background: Cloning techniques are applied to an endangered orchid species in order to reproduce individual plants and to preserve their genetic characteristics. Objectives: To establish a new protocol for clonal propagation of a threatened orchid of horticultural importance. Study species: Trichocentrum stramineum, a threatened orchid endemic to Mexico. Study site and duration: Totutla, Veracruz, Mexico. All experiments were designed and carried out at the Botanical Garden-UNAM and the IIAF-UMSNH over a course of six years. Methods: Seeds were germinated in a modified KC basal medium; protocorms and apical bud explants were obtained from the resulting in vitro plants and cultivated with or without plant growth regulators (PGRs). Both experimental groups were subcultured in order to evaluate the number of protocorm-like bodies (PLBs) and buds per explant. Results: On average, protocorms generated 51.2 and 54.1 PLBs in the absence or presence of 1 mg l-1 6-benzyladenine (BA), respectively, while 13.1 and up to 23.7 PLBs and / or shoots were observed on the apical bud explants in the absence or presence of 1 mg l-1 kinetin, respectively. In both cases, responses were direct, without the formation of an intervening callus. Approximately 200 PLBs were subcultured and developed into whole plants within 14 weeks. These were acclimatized to greenhouse conditions with a 90 % survival rate after 12 weeks. After 44 weeks, flowering was observed (3 %) individuals measuring at least 12 cm in height. Conclusions: The developed protocol proved to hold great potential for commercial propagation and conservation programs.

<p align="justify"><em>Prosthechea sp</em>. es una orquídea silvestre apetecida por el color y forma de sus flores, que está desapareciendo paulatinamente en el municipio de Fusagasugá (Cundinamarca, Colombia) a causa de la deforestación y extracción por parte de la población presente en la zona rural. Se probó el efecto de cuatro concentraciones (0; 1,5; 3 y 4 mg/l-1) de ácido naftalenacético (ANA) sobre el desarrollo de protocormos de Prosthechea sp. El cultivo se llevó a cabo bajo condiciones in vitro durante 42 semanas, de manera asimbiótica. El medio de enraizamiento suplementado con 1,5 mg/l-1 de ANA tuvo un efecto nulo sobre la formación de raíces, sin embargo, se presentó proliferación de protocormos por gemación adventicia en un 80 %. Las concentraciones de 3 y 4 mg/l-1 de ANA obtuvieron desarrollos de plántulas en 70 y 65 % respectivamente, al presentar abundantes raíces gruesas y pelos radicales. Se observó la respuesta triple en los tratamientos con mayor concentración de ANA debido a la síntesis de etileno, afectando el geotropismo de la raíz. Esta investigación servirá como base para futuros proyectos de multiplicación y conservación, contribuyendo con la preservación de la flora silvestre y los recursos genéticos con gran potencial ecológico y económico como lo son las orquídeas.</p>

Laelia halbingeriana Salazar & Soto Arenas is a wild epiphytic orchid endemic to Oaxaca, Mexico, that is collected with no plans for its management or conservation. The objective of this study was to propagate and evaluate the in vitro development of this species in the culture media with a variety of mineral salts and growth regulators. Groups of three plants, 1.2 to 1.4 cm tall, were established in receptacles with culture medium containing 30 g L-1 sucrose, 1 mg L-1 thiamine, 100 mg L-1 inositol and 0.5 mg L-1 benzyl aminopurine (BAP). Proportions of Murashige and Skoog (1962) or Knudson (1946) mineral salts were varied (100, 66 and 33%). The growth regulators indoleacetic acid (IAA) and kinetin (Kin) were used at various concentrations (0.5 + 0.5 mg L-1 , 1 + 1 mg L-1 and 2 + 2 mg L-1). pH was adjusted to 5.8, and 5.5 g L-1 agar was added. After 120 days of incubation, the plants that were cultivated in 66% and 100% Knudson media were 2.1 cm tall and had 4.8 roots, while the plants in 100% Murashige and Skoog media were 1.5 cm tall and had 2.5 roots. For 120 days, the plants had a logarithmic trend in vertical growth (R 2 ≥0.96). Several plant characteristics were evaluated simultaneously using a cluster analysis, which found that the best culture medium for Laelia halbingeriana development contains 100% Knudson mineral salts, 0.5 mg L-1 BAP, 0.5 mg L-1 IAA, and 0.5 mg L-1 KIN.
