Properties of memory T lymphocytes isolated from the mixed leukocyte reaction.

Abstract: During the primary mixed leukocyte reaction, T lymphocytes of the lyt-2-helper subclass proliferate in response to transplantation antigens on allogeneic dendritic cells. We have isolated populations of antigen-specific proliferating lymphoblasts and recultured them in fresh medium. Within 2 days, the blasts become smaller in size, lose responsiveness to T-cell growth factor or interleukin 2, but retain vigorous reactivity to the original alloantigen. Two new biologic properties of these "memory" lymphocytes h… Show more

“…Lymphoblasts were sensitized to lectin (3 ~,g/ml Con A, Pharmacia Fine Chemicals, Piscataway, N J), alloantigens, or soluble proteins (50 ~g/ml of either KLH [Calbiochem-Behring, San Diego, CA] or OVA [Sigma Chemical Co., St. Louis, MO]). The procedure was as described (4)(5)(6), and the cell yields are summarized in Table I. Briefly, 5 x 104 spleen DC were cultured with 5 × 106 T cells in 1 ml medium (RPMI-1640 supplemented with 10% FCS 20,,g/ml gentamicin, and 5 x l0 -~ M 2-ME) in 16-mm-diam culture wells.…”

“…Memory T cells were prepared by resting the blasts for three or more days in the absence of exogenous IL-2 or alloantigen (19) . The blasts and memory populations were >70% allospecific, as assessed in rapid DC-T binding assays (28 Mixtures of accessory-cell depleted B and T lymphocytes were obtained by passing spleen suspensions over Sephadex G10 (17) .…”

“…Although the clusters contain < 10% of the available T cells, these immunocompetent units are responsible for almost all the interleukins and growth factors produced in the cultures (3,8,14) and physical blocking of cluster formation by interposing an artificial membrane abrogates the observed immunological responses (15). In all instances the responsible cell is an Lyt-2-helper T cell (4,5); but murine DC can also cluster with and activate cytotoxic T cells (1 6).…”

“…generate active helper cells from unprimed and memory lymphocytes, and cause B cells to secrete antibodies (2)(3)(4)(5). Cultured spleen cells depleted of DC failed to develop anti-TNP cytotoxic T cells while the addition of a small number of DC restored the antigen dependent response (6).…”
