Twenty-one renal cell carcinomas were analyzed for the presence of androgen progestin and estrogen receptors by agar gel electrophoresis and a dextran-coated charcoal assay. An androgen receptor could be demonstrated in 3 tumors, a progestin or estrogen receptor was not detectable. It was therefore concluded that renal cell carcinoma is generally not a hormone-dependent tumor, and a hormonal treatment of metastatic renal cell carcinoma as first-line therapy does not seem to be justified.

The content of cytoplasmic androgen receptors (Bmax) was analyzed in non-tumoral renal tissue of 12 men and 15 postmenopausal women using a synthetic androgen (methyltrienolone) as ligand and a method of dextran-coated charcoal. The Bmax in both sexes was compared, establishing correlations between it and circulating levels of testosterone, dehydroepiandrosterone sulfate, androstenedione, and estradiol to find out the possible influence of the hormonal environment on androgen receptors in the human kidney. No differences in Bmax were observed between males (46.3 ± 24 fmol/g of tissue) and females (45.4 ± 26 fmol/g), in spite of the significantly greater (p < 0.01) levels of circulating testosterone in the former group. No significant linear correlations existed between any of the steroids analyzed and the Bmax. These results demonstrate the existence of androgenic receptors in non-tumoral human kidney and indicate that its content is not regulated by circulating levels of testosterone. The concentrations of the principal extratesticular androgens and estradiol do not seem to have a quantitative influence on these androphilic proteins either.

Summary: In normal human kidney from adult males cytoplasmatic components which bound 17|3-oestradiol specifically and with high affinity were demonstrated by dextran-coated charcoal assay, sucrose gradient centrifugation and agar gel electrophoresis. The dissociation constant of the oestradiol-binder complex amounted to 2.2 ± 0.1 9 mol/1. The binding capacity was limited to 34.0 ± 9.7 fmol/mg of cytosol protein. Sedimentation in sucrose gradient revealed the bulk of these components to be in the 4 S region. The binding entities could be clearly separated from sex hormone-binding globulin by agar gel electrophoresis. The ligand specificity for binding to these components indicated a requirement for oestrogens. The fact that an excess of aldosterone had no competitive effect on oestradiol binding suggests that the oestrogen-binding sites are independent of mineralocorticoid receptors. It is concluded that the specific binding components in human kidney have the properties of oestrogen receptors. Identifizierung und teilweise Charakterisierung spezifisch östrogenbindender Komponenten in der menschlichen NiereZusammenfassung: In normalen menschlichen Nieren erwachsener männlicher Patienten wurden mittels Kohle-Adsorptionstechnik, Sucrose-Dichtegradientenzentrifugation und Agargelelektrophorese zytoplasmatische Komponenten nachgewiesen, die 17j3-Östradiol spezifisch und mit hoher Affinität binden. Die Dissoziationskonstante des Östradiol-Binderkomplexes betrug 2,2 ± 0,1 X l Cf 9 mol/1. Die Bindungskapazität war begrenzt und belief sich auf 34,0 ± 9,7 fmol/mg Cytosolprotein. Bei der Sücrose-Dichtegradientenzentrifugation sedimentierte der größte Teil dieser Komponenten in der 4S-Region. Durch Agargelelektrophorese konnten diese Komponenten klar von sexualhormonbindendem Globulin getrennt werden. In Kompetitionsexperimenten konnte gezeigt werden, daß diese Komponenten spezifisch Östrogene binden. Da ein Überschuß an Aldosteron keinen kompetitiven Effekt auf die Bindung von 17/J-Östradiol hat, sind die östrpgenbindenden Komponenten nicht mit Mineralcorticoidrezeptoren identisch. Die Untersuchungen zeigen, daß die östrOgeribindenderi Komponenten die Eigenschaften von Östrpgenrezeptoren aufweisen.
