Prophylaxis of Malaria

Abstract: Malaria prevention in travelers to endemic areas remains dependent principally on chemoprophylaxis. Although malaria chemoprophylaxis refers to all malaria species, a distinction should be drawn between falciparum malaria prophylaxis and the prophylaxis of the relapsing malaria species (vivax & ovale). While the emergence of drug resistant strains, as well as the costs and adverse reactions to medications, complicate falciparum prophylaxis use, there are virtually no drugs available for vivax prophylaxis, besi… Show more

“…Furthermore, ELQ‐400 was evaluated in a causal prophylaxis assay and found to be capable of inhibiting LS infection by firefly luciferase‐expressing P. yoelii (PyLuc) sporozoites . LS (causal) prophylaxis refers to drugs that are active against malaria parasites within the hepatocytes, that is, PQ and malarone . Remarkably, a single dose of ELQ‐400 when administered orally 1 hr after sporozoite inoculation completely prevented the development of both LS and blood‐stage infections at oral doses as low as 0.08 mg/kg.…”

“…The most commonly recommended regimens of chemoprophylaxis are as follows: doxycycline 100 mg once daily (started one day before traveling, and continued for four weeks after returning); mefloquine 250 mg once weekly (started 2.5 weeks before traveling, and continued for four weeks after returning); atovaquone/proguanil one tablet daily (started one day before traveling, and continued for one week after returning) [27].…”
