Protection of silver nanoparticles using Eysenhardtia polystachya in peroxide-induced pancreatic β-Cell damage and their antidiabetic properties in zebrafish

Abstract: BackgroundThe aim was to explore the efficacy of extract of Eysenhardtia polystachya-loaded silver nanoparticles (EP/AgNPs) on pancreatic β cells, INS-1 cells, and zebrafish as a valuable model for the study of diabetes mellitus.Materials and methodsEP/AgNPs were synthesized using methanol/water bark extract of E. polystachya and characterized using various physicochemical techniques.ResultsImmersion of adult zebrafish in 111 mM glucose solution resulted in a sustained hyperglycemic, hyperlipidemic state, and … Show more

“…The nanoformulation elevated pancreatic β cells survival and ameliorated insulin resistance and hyperglycemia, as well as dyslipidemia in glucose-induced diabetes in zebrafish. Also, in INS1 pancreatic β cell line intoxicated with hydrogen peroxide (H 2 O 2 ), nanoformulated extract could significantly restore the insulin secretory ability of cells, which indicates antidiabetic activity of extract to be, at least in part, attributed to its antioxidant properties (Campoy et al, 2018).…”

“…The synthesis was confirmed by UV-visible spectroscopy, with peaks obtained at 409 and 413 nm for AgNP-TS and AgNP-T, respectively (Fig. 1A), indicating the presence of elemental silver 35,36 . Analysis of the filtrates used in the synthesis revealed peaks at 221 and 215 nm for the samples obtained with and without stimulation by the cell wall of S .…”

“…The zebrafish has been used as an experimental model due to the great physiological similarity that it presents with mammals, in addition to having a small body size, low cost, and large clutch size [ 39 ]. Therefore, this model presents significant potential for pharmacological and toxicological studies.…”
