Protective effect of decursin and decursinol angelate-rich Angelica gigas Nakai extract on dextran sulfate sodium-induced murine ulcerative colitis

Abstract: The findings of this study verified that AGNE significantly improves clinical symptoms and reduces the activity of various inflammatory mediators. These results indicate the AGNE has the therapeutic potential in mice with DSS-induced UC.

“…In UC pathogenesis, increased secretions of the pro-inflammatory cytokines IL-6, TNF-α, and IFN-γ are closely implicated with intestinal inflammatory responses. IL-6 elevates the body temperature by inducing production of acute-phase response proteins, accompanied with the infiltration of inflammatory immune cells and increased permeability of vascular endothelial cells by TNF-α [13]. The clinical application of TNF-α and IL-6 antagonists (e.g., infliximab, adalimumab, and tocilizumab) demonstrated therapeutic effects against acute and chronic UC.…”

“…5 We were unable to identify which herbs the patient ingested; however, Angelica gigas Nakai, Cnidium officinale Makino, Citrus unshiu Markovich, and Glycyrrhiza uralensis Fischer are thought to be useful for anemia in Korea, so it can be speculated that the patient could have ingested these herbs. Angelica gigas Nakai is known to reduce cardiac contraction, 8 but there are no reports that it causes arrhythmia; the three other herbs also lack evidence of cardiac toxicity. Therefore, she might have taken additional herbs and one or more of them could have caused the refractory VT.…”

“…An acute toxicity test of AGNE determined that the LD50 was >2000 mg/kg. The dosage used in our experiment was determined based on the dosage verified in a previous study of the positive effects of AGNE on ulcerative colitis [ 19 , 20 ]. Also, the dosage was within the range in which terfenadine does not cause adverse reactions [ 21 ].…”
