Protective Effect of Grape Seed Procyanidins against H2O2‐Induced Oxidative Stress in PC‐12 Neuroblastoma Cells: Structure–Activity Relationships

Abstract: This work evaluated the protective effect of procyanidins against oxidative stress in PC-12 neuroblastoma cells and established their activity-structure relationships, which provides useful cellular evidence for the further investigating the structure-optimizing and function-exploiting of procyanidins.

“…This property was reported for polyphenols present in wine, such as resveratrol, that was shown to inhibit cyclooxygenase 2 (COX-2) and decrease prostaglandin E 2 (PGE 2 ) in the presence of beta-amyloid peptide [69]. Moreover, procyanidins from seed grapes showed antioxidant benefits in PC-12 cells [70]. Our results show that dark beer is the treatment that presents the highest antioxidative effect, at 30 min and 24 h, and is able to produce the highest increase of adenosine A 1 receptor protein levels in both cells models analyzed.…”

“…The phenols in Vitis vinifera L. seeds provide health benefits and proanthocyanidins, also known as condensed tannins, play a potential nutraceutical role. The antioxidant and free radical scavenging abilities of phenols have been demonstrated both in vivo and in vitro, protecting more than vitamins E or C and β-carotene [ 42 , 43 , 44 ]. Other activities—such as anti-inflammatory [ 45 ], antimicrobial [ 46 ], antiulcer [ 47 ], and anticancer—have also been reported [ 48 , 49 , 50 ].…”

“…Procyanidins, the water-soluble and fat-soluble compounds, are the most abundant polyphenols found in foods such as fruits, red wine, green tea (C adiz-Gurrea et al 2014) and in bark and the seeds of many plants (Luo et al 2018). Procyanidins arise from flavan-3-ol units, including catechin and epicatechin, and exist in a range of forms (dimers, trimers, tetramers, pentamers, and polymers).…”

“…Among all types of procyanidins, the dimers are the most abundant in nature and have stronger antioxidant activities. Procyanidins also have anti-inflammatory (Youn et al 2017), antioxidant (Luo et al 2018), and hypoglycaemic properties (Gonzalez-Abuin et al 2015). Therefore, they are used in the prevention and treatment of atherosclerosis, coronary heart diseases (Goszcz et al 2017), DM (Gonzalez-Abuin et al 2015;Ogura et al 2016), and cancers (Choy et al 2016).…”
