ARTICLE INFO ______________________________________________________________ ______________________Objective: To analyze the oxidative damage and histopathological alterations caused by ischemia-reperfusion (I/R) injury and ameliorative effects of carvedilol (CVD) in the rat testis. Materials and Methods: Twenty-one male rats were randomized into 3 groups as follows: Group I (n = 7); control (sham) group, Group II (n = 7); I/R group, in which I/R injury was performed by torsing the left testis 720º clockwise for 2 hours and detorsing for 2 hours. Group III (n = 7); CVD treatment group; in addition to I/R process, one--dose of CVD was administered (2mg/kg, i.p) 30 min. before detorsion. Levels of antioxidant enzymes, superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) and levels of malondialdehyde (MDA) and protein carbonyl (PC) were determined in testicular tissues and serum of rats. Testicular tissues were also examined histopathologically and Johnsen scores were determined. Results: Activities of SOD and GSH-Px in serum and testicular tissues were increased by I/R, but administration of CVD decreased these levels (p < 0.001 and p = 0.001).Significantly increased MDA levels in serum and testicular tissues were decreased by CVD treatment (p < 0.001 and p = 0.001). Concerning PC levels in serum and testicular tissues, there was no statistically significant difference between the groups (p = 0.989 and p = 0.428). There was not a statistically significant difference in terms of mean Johnsen scores between the groups (p = 0.161).Conclusions: Administration of CVD decreased oxidative damage biochemically in the rat testis caused by I/R injury, but histopathologically no change was observed between all of the groups.

Objectives: To investigate the protective role of thymoquinone (TQ) on unilateral testicular ischemia-reperfusion (I/R) injury in mice. Materials and Methods: Experiments were performed on male C57BL/6 mice (8 weeks old, 20–25 g). The animals were divided into 3 groups including 6 mice in each group: control (sham), torsion/detorsion (TD) and TD+TQ. Mice, except the sham-operated group, were subjected to left unilateral torsion (720° rotation in the clockwise direction). The experiments were finished after sham operation time for controls, 120 min torsion and 240 min detorsion for the other groups. In the TD+TQ group 10 mg TQ was injected intraperitoneally 30 min before detorsion. Results: In the TD group total oxidative stress (TOS), oxidative stress index (OSI) and malondialdehyde (MDA) levels were higher than in the controls. TQ treatment decreased MDA, TOS and OSI values, but did not affect the total antioxidant capacity and myeloperoxidase activity in the TD+TQ group. Upon histological examination, mice in the TD group displayed moderate-to-severe disruption of the seminiferous epithelium. Treatment with TQ resulted in significantly reduced histological damage associated with I/R injury. Conclusion: Our results suggested that TQ treatment may have a protective effect on testicular I/R injury.

Amaç. Testis torsiyonu, iskemi reperfüzyon hasarı formunda önemli bir akut skrotum nedenidir. Başarılı ve erken bir detorsiyon yöntemi ile morbidite azaltılabilir. Bu çalışmada deneysel tek taraflı testis torsiyonu ve detorsiyonu modelinde N-asetilsistein'in her iki testise olan etkisinin araştırılması amaçlandı. Yöntem. Çalışmada 32 adet prepubertal erkek Wistar albino sıçan kullanıldı. Sıçanlar malondialdehid düzeyi ve histopatolojik hasar araştırılmak üzere kontrol, torsiyon, detorsiyon ve detorsiyon/N-asetilsistein gruplarına ayrıldı. Kontrol grubunda yer alan deneklere torsiyon işlemi uygulanmadan testisleri alındı. Diğer gruplardaki deneklerin sağ testislerine dörder saatlik torsiyon işlemi uygulandı. Detorsiyon/N-asetilsistein grubunda detorsiyondan 15 dk önce intravenöz N-asetilsistein verildi. Detorsiyon ve detorsiyon/Nasetilsistein gruplarında reperfüzyon sonrası dört saat daha beklendi. Deney sonunda tüm gruplarda her iki testis de çıkarılarak malondialdehid düzeyleri çalışıldı ve histopatolojik olarak değerlendirildi. Malondialdehid sonuçları Kruskal-Wallis varyans analizi ve Mann Whitney U testleriyle değerlendirildi. Farkın p için 0,05'den küçük olduğu değerler anlamlı kabul edildi. Bulgular. Her iki testise ait malondialdehid düzeyleri tüm gruplarda kontrol grubuna göre daha yüksek olarak bulundu (p<0,05). Detorsiyon/N-asetilsistein grubu ile detorsiyon grubu arasında her iki testis dokusuna ait malondialdehid düzeyleri bakımından anlamlı fark saptanmadı (p>0,05). İşlem sonrasında kontrol grubu dışındaki tüm grupların torsiyone testislerinde interstisyel hemoraji ile seminifer tübül yapılarında düzensizlik gözlendi. Karşı tarafta interstisyel ödem ve bazı olgularda germ hücrelerinin dökülmesi dışında normal histoloji izlendi. Sonuç. Sonuç olarak oluşturduğumuz deneysel testis torsiyonu modelinde detorsiyondan önce intravenöz N-asetilsistein uygulamasının her iki testiste malondialdehid düzeyleri ve histopatolojik hasar üzerinde bir etkisinin olmadığı saptandı. In this study, 32 prepubertal male albino Wistar rats were used. The rats were divided into four groups as control, torsion, detorsion and detorsion/N-acetylcysteine in order to determine the malondialdehyde level and histopathological damage. Rats in the control group were killed after the testes were removed without torsion. Right testes of the rats underwent four hours of torsion in other groups.
