SummaryIntroduction: Baikal skullcap (Scutellaria baicalensis Georgi) is an important medicinal plant, indigenous to Asia. Due to a wide range of pharmacological activities, its roots has been used for ages in Traditional Chinese Medicine. Recently, the species has become an object of interest of Western medicine, as well. Objective: The aim of the study was to determine the variability of Baikal skullcap population originated from Mongolia and cultivated in Poland, in terms of content and composition of flavonoids in the roots. Methods: The objects of the study were 15 individual plants, selected within examined population and cloned in order to obtain a sufficient amount of raw material. The total content of flavonoids in roots was determined according to Polish Pharmacopeia 6th. The qualitative analysis of flavonoids was carried out using HPLC, Shimadzu chromatograph. Results: The dry mass of roots ranged from 25.88 to 56.14 g × plant-1. The total content of flavonoids (expressed as a quercetin equivalent) varied between 0.17 and 0.52% dry matter (DM). Nine compounds were detected within the group, with oroxylin A 7-Oglucuronide (346.90-1063.00 mg × 100 g-1DM) as a dominant, which differentiated investigated clones at the highest degree (CV=0.27). Baicalin (391.40-942.00 mg × 100 g-1DM), wogonoside (324.00-641.10 mg × 100 g-1DM) and hesperetine 7-O-glucoside (163.00-346.32 mg × 100 g-1DM) were also present in a considerable amounts. Clone 7 was distinguished by the highest content of all investigated compounds, except wogonin and oroxylin A 7-O-glucuronide. Conclusions: Results obtained in present study show a high variability within Baical skullcap investigated population in respect of flavonoid compounds detected in roots. Thus, the results may be used in future investigations concerning the selection and breeding of this species.

Aim: To study the metabolic and pharmacokinetic profile of scutellarin, an active component from the medical plant Erigeron breviscapus (Vant) Hand-Mazz, and to investigate the mechanisms underlying the low bioavailability of scutellarin though oral or intravenous administration in rats. Methods: HPLC method was developed for simultaneous detection of scutellarin and scutellarein (the aglycone of scutellarin) in rat plasma, urine and feces. The in vitro metabolic stability study was carried out in rat liver microsomes from different genders. Results: After a single oral dose of scutellarin (400 mg/kg), the plasma concentrations of scutellarin and scutellarein in female rats were significantly higher than in male ones. Between the female and male rats, significant differences in AUC, t max2 and C max2 for scutellarin were found. The pharmacokinetic parameters of scutellarin in the urine also showed significant gender differences. After a single oral dose of scutellarin (400 mg/kg), the total percentage excretion of scutellarein in male and female rats was 16.5% and 8.61%, respectively. The total percentage excretion of scutellarin and scutellarein in the feces was higher with oral administration than with intravenous administration. The in vitro t 1/2 and CL int value for scutellarin in male rats was significantly higher than that in female rats. Conclusion:The results suggest that a large amount of ingested scutellarin was metabolized into scutellarein in the gastrointestinal tract and then excreted with the feces, leading to the extremely low oral bioavailability of scutellarin. The gender differences of pharmacokinetic parameters of scutellarin and scutellarein are due to the higher CL int and lower absorption in male rats.

Breviscapine ameliorates hypertrophy of cardiomyocytes induced by high glucose in diabetic rats via the PKC signaling pathway

Aim: To investigate the influence of breviscapine on high glucose-induced hypertrophy of cardiomyocytes and the relevant mechanism in vitro and in vivo. Methods: Cultured neonatal cardiomyocytes were divided into i) control; ii) high glucose concentrations; iii) high glucose+PKC inhibitor Ro-31-8220; iv) high glucose+breviscapine; or v) high glucose+NF-κB inhibitor BAY11-7082. Cellular contraction frequency and volumes were measured; the expression of protein kinase C (PKC), NF-κB, TNF-α, and c-fos were assessed by Western blot or reverse transcription-polymerase chain reaction (RT-PCR). Diabetic rats were induced by a single intraperitoneal injection of streptozotocin, and randomly divided into i) control rats; ii) diabetic rats; or iii) diabetic rats administered with breviscapine (10 or 25 mg·kg -1 ·d -1 ). After treatment with breviscapine for six weeks, the echocardiographic parameters were measured. All rats were then sacrificed and heart tissue was obtained for microscopy. The expression patterns of PKC, NF-κB, TNF-α, and c-fos were measured by Western blot or RT-PCR. Results: Cardiomyocytes cultured in a high concentration of glucose showed an increased pulsatile frequency and cellular volume, as well as a higher expression of PKC, NF-κB, TNF-α, and c-fos compared with the control group. Breviscapine could partly prevent these changes. Diabetic rats showed relative cardiac hypertrophy and a higher expression of PKC, NF-κB, TNF-α, and c-fos; treatment with breviscapine could ameliorate these changes in diabetic cardiomyopathy. Conclusion: Breviscapine prevented cardiac hypertrophy in diabetic rats by inhibiting the expression of PKC, which may have a protective effect in the pathogenesis of diabetic cardiomyopathy via the PKC/NF-κB/c-fos signal transduction pathway.
