Protein interaction mapping: ADrosophilacase study

Abstract: The Drosophila (fruit fly) model system has been instrumental in our current understanding of human biology, development, and diseases. Here, we used a high-throughput yeast two-hybrid (Y2H)-based technology to screen 102 bait proteins from Drosophila melanogaster, most of them orthologous to human cancer-related and/or signaling proteins, against high-complexity fly cDNA libraries. More than 2300 protein-protein interactions (PPI) were identified, of which 710 are of high confidence. The computation of a reli… Show more

“…After selection on medium lacking Leu, Trp, and His, seven positive clones were picked, and the corresponding prey fragments were amplified by PCR and sequenced at their 5# and 3# junctions. Sequences were then filtered and contiged as described previously (Formstecher et al, 2005) and compared with the latest release of the GenBank database using BLASTN (Altschul et al, 1997). A predicted biological score (PBS) was applied to assess the reliability of each interaction, as described previously (Formstecher et al, 2005).…”

“…In the absence of Bitesize function, early aspects of apical-basal polarity, including the formation of adherens junctions, occur normally but, subsequently, epithelial organization and the adherens junctions break down [34,35] . Loss of moesin function display a disruption in overall morphology and epithelial integrity [36,37] .…”
