Protein profiling in pathology: Analysis and evaluation of 239 frozen tissue biopsies for diagnosis of B‐cell lymphomas

Abstract: SELDI-TOF MS protein profiling allows for reliable identification of the majority of malignant lymphoma cases; however, further validation and testing robustness in a diagnostic setting is needed.

“…Four [26][27][28][29] included kinds of stages and subtypes of lymphoma. One [20] focused on B-cell lymphomas. The remaining three papers were related to Coetaneous T-cell lymphomas (CTCL) [30], mycosis fungoides (MF) [19] and diffuse large-B-cell lymphoma (DLBCL) [31], respectively.…”

“…Seven studies clearly pointed out that treatment was withheld until both the index test and reference standard were performed except Jiang [28], Cheng [27], Escher [30] and Escher [19], which were not available. Four studies, Jiang [28], Chen [25], Jansen [20] and Zhang [24], were prespecified objectively, and remains were not available.…”

“…It is a more difficult to standardize compared with DNA and even RNA approaches. Jansen et al [53] determined the potential value of protein profiling of tissue samples by assessing how precise this approach enables discrimination of B cell lymphoma from reactive lymph nodes and how well the profiles can be used for lymphoma classification. Protein lysates from lymph nodes (n=239) from patients with the diagnosis of reactive hyperplasia (n=44), FL (n=63), DLBL (n=43), MCL (n=47), and CLL (n=42) were analyzed by SELDI-TOF MS.…”
