Proteolytic activation of the epithelial sodium channel (ENaC) by the cysteine protease cathepsin-S

Abstract: Proteolytic processing of the amiloride-sensitive epithelial sodium channel (ENaC) by serine proteases is known to be important for channel activation. Inappropriate ENaC activation by proteases may contribute to the pathophysiology of cystic fibrosis and could be involved in sodium retention and the pathogenesis of arterial hypertension in the context of renal disease. We hypothesized that in addition to serine proteases, cathepsin proteases may activate ENaC. Cathepsin proteases belong to the group of cystei… Show more

“…Defolliculated stage V-VI oocytes were injected (Nanoject II automatic injector, Drummond) with 0.5 ng of TRPV4 cRNA alone, 10 ng of PAR 2 cRNA alone, or both TRPV4 and PAR 2 cRNA. Oocytes were studied 2 days after injection using the two-electrode voltage-clamp technique as described (37,38,44). A Ca 2ϩ -free solution was used to prevent activation of endogenous Ca 2ϩ -activated chloride channels by TRPV4-mediated Ca 2ϩ influx and to delay a Ca 2ϩ -induced decay of TRPV4 current (31,45).…”
