Objective: Autism spectrum disorder is a neurodevelopmental disease in which impaired social behaviors, impaired sociality, and restricted and repetitive behaviors are seen. Bumetanide is a loop diuretic that inhibits Na + –K + –2Cl − cotransporter 1 and it is currently used in clinical phase studies in patients with autism spectrum disorder. In present research, it is purposed to demonstrate the beneficial effects of torasemide which is another Na + –K + –2Cl − cotransporter 1 inhibitor on an experimental autism model induced with propionic acid by providing imaging and brain tissue investigations. Methods: Male Wistar rats were used in the present study (n = 30). Propionic acid of 250 mg/kg/day was administrated intraperitoneally in rats to induce autism for 5 days. Three groups were created for present study as follows: group 1, normal control (n = 10); group 2, propionic acid and saline given group (n = 10); group 3, propionic acid + tora-semide-administrated group (n = 10). Results: Torasemide group scored higher on behavioral tests compared to saline group. The brain levels of malondialdehyde, tumor necrosis factor-alpha, interleukin-2, interleukin-17, and Nuclear Factor kappa B (NF-κB), Glial fibrillary acidic protein (GFAP) were remarkably higher in propionic acid + saline group. In histopathology assessments, torasemide group had higher neuronal count of Cornu Ammonis 1, neuronal count of Cornu Ammonis 2 in hippocampus, and Purkinje cells in cerebellum. GFAP immunostaining index (Cornu Ammonis 1) and cerebellum were lower in torasemide group. Magnetic resonance spectroscopy revealed that mean lactate value was higher in propionic acid + saline group compared to torasemide group. Conclusion: Our experimental results showed that torasemide might enhance gamma-aminobutyric acid activity. Torasemide can be considered another promising Na + –K + –2Cl − cotransporter 1 inhibitor in the treatment of autism with a longer half-life and less side effects after further studies.

Iron is an essential trace element to maintain the normal physiological function of organisms. In this study, the urine proteome of rats before and after short-term intragastric administration of polysaccharide-iron complex (28mg/kg/d iron, which is equivalent to the dose of anemia prevention in adults) was compared and analyzed by using two analysis methods: individual comparison and group comparison. Many different proteins were reported to be related to iron, including 2', 3' -cyclic nucleotide 3' -phosphodiesterase (CNPase) (7.7 times higher than that after gavage, p=0.0039), p38 (14.5 times higher than that before gavage, p=0.003), etc. In the individual comparison, Hepcidin was up-regulated in 4 rats simultaneously. The biological processes of differential protein enrichment include carbohydrate metabolism, iron ion reaction, apoptosis regulation, hematopoietic progenitor cell differentiation, etc. Molecular functions (e.g., complement binding, hemoglobin binding, etc.), KEGG pathways (e.g., complement and coagulation cascade, cholesterol metabolism, malaria, etc.) have also been shown to be associated with iron. This study contributes to the in-depth understanding of the biological function of iron from the perspective of urine proteomics, and provides a new research perspective for the prevention, diagnosis, treatment and monitoring of iron-related disorders.

Background: Loop diuretics are commonly used for patients with heart failure (HF) but it remains unknown if one loop diuretic is clinically superior.Hypothesis: Biomarkers and proteomics provide insight to how different loop diuretics may differentially affect outcomes.Methods: Blood and urine were collected from outpatients with HF who were taking torsemide or furosemide for >30 days. Differences were assessed in cardiac, renal, and inflammatory biomarkers and soluble protein panels using the Olink Cardiovascular III and inflammation panels.Results: Of 78 subjects, 55 (71%) were treated with furosemide and 23 (29%) with torsemide, and 25 provided a urine sample (15 treated with furosemide, 10 with torsemide). Patients taking torsemide were older (68 vs 64 years) with a lower mean eGFR (46 vs 54 ml/min/1.73 m 2 ), a higher proportion were women (39% vs 24%) and Black (43% vs 27%). In plasma, levels of hs-cTnT, NT-proBNP, and hsCRP were not significantly different between groups. In urine, there were significant differences in urinary albumin, β-2M, and NGAL, with higher levels in the torsemide-treated patients. Of 184 proteins testing in Olink panels, in plasma, 156 (85%) were higher in patients taking torsemide but none were significantly different after correcting for false discovery.Conclusions: We show differences in urinary biomarkers but few differences in plasma biomarkers among HF patients on different loop diuretics. Olink technology can detect differences in plasma protein levels from multiple biologic domains. These findings raise the importance of defining differences in mechanisms of action of each diuretic in an appropriately powered study.
