Proteomics integrated with metabolomics: analysis of the internal causes of nutrient changes in alfalfa at different growth stages

Abstract: BackgroundAlfalfa (Medicago sativa L.) is one of the most important forage resources in the world due to its high nutritive value. However, its nutritional quality decreases during the transition from budding to flowering. Previous research revealed a decreased crude protein content and increased fibre content in alfalfa forage harvested at later maturity stages, leading to a reduction in nutritional quality. However, the reasons for this phenomenon have not been explained at the molecular level.ResultsIn this… Show more

“…By using XCMS software, the nonlinear alignment, automatic integration, extraction of the peak intensities, peak alignment, and data filtering were finished. All the metabolites were identified by accuracy mass (< 25 ppm) matching and secondary mass spectrogram matching, and the score cutoff was set as 0.8 [31]. MetaboAnalyst 4.0 was used for statistical analysis [32].…”

“…using a UHPLC (1290 Infinity LC, Agilent Technologies) coupled to a quadrupole time-of-flight (AB Sciex TripleTOF 6600) in Shanghai Applied Protein Technology Co., Ltd. According to Wenqiang Fan's method, the HILIC separation was carried out samples were analyzed by using a 2.1 mm × 100 mm ACQUIY UPLC BEH 1.7 µm column (waters, Ireland) 69 . In the ESI positive and negative mode, the mobile phase contained A = 25 mM ammonium acetate and 25 mM aqueous ammonium hydroxide, B = acetonitrile.…”

“…Alanine, purines and pyrimidines content was reduced before the protein synthesis, which could lead to protein synthesis decreased, which was the reason of CP content in solid stage is lower than the jointing stage. The content of phenylalanine increased, but the protein content did not increase in the related metabolic pathway [18]. It can be considered that phenylalanine mainly comes from the hydrolysis of protein, which may also be one of the reasons for the decrease of protein content.…”
