Prothrombin G20210A Mutation: Is It Associated with Pre-Eclampsia?

Abstract: Objective: A strong independent association between the prothrombin G20210A gene mutation and pre-eclampsia has been reported in an Italian population. This result was not confirmed in a subsequent study in a Dutch population. The objective of this study was to further test the hypothesis that the prothrombin G20210A mutation is associated with pre-eclampsia/eclampsia. Methods: Seventeen eclamptics and 67 pre-eclamptics were recruited from 34 multicase Australian/New Zealand families. An additional 105 unrelat… Show more

Diagnosis of PE and assessment of its severity were in strict accordance with the American College of Obstetricians and Gynecologists criteria.…”

“…In addition, the limited size of studies of the association of thrombophilia with PE reduces their statistical significance and may result in its overestimation or underestimation. 15,17,23,24,27,31,32 The size of our sample is one of the strengths of our study. Even so, it was insufficient to detect significant differences in the prevalence of inherited deficiency of protein C, protein S, and antithrombin III, all rather infrequent.…”

“…Many studies have now been conducted investigating possible associations between PE/E and candidate genes, including angiotensinogen [43,44], methylenetetrahydrofolate reductase [23,45], prothrombin [21], factor V Leiden [46] and others. These studies have failed to reach agreement with one group reporting association of a particular candidate gene with PE/E and another group unable to confirm the association.…”

“…No difference in homozygosity for MTHFR was found between the 2 groups (preeclampsia 3% vs controls 6%). Higgins et al [62], tested the prothrombin mutation in 13 eclamptic and 74 preeclamptic women with severe disease coming from 34 families of mostly Anglo-Saxon origin. They were compared to 119 controls.…”
