Puerarin Accelerates Peripheral Nerve Regeneration

Abstract: This study investigates the effect of puerarin (PR) on peripheral nerve regeneration in vitro and in vivo. PR at concentrations of 1, 10, and 100 μM significantly promoted survival and outgrowth of cultured Schwann cells, as compared to the controls treated with culture medium only. in vivo study, peripheral nerve regeneration was evaluated across a 15-mm gap in the sciatic nerve of rats using a silicone rubber nerve chamber filled with PR solution. The control group chambers were filled with normal saline onl… Show more

“…Puerarin had an antagonistic action on LPS-activated N9 microglia cells and might be beneficial to the treatment of neurodegenerative disorders (Bai et al, 2010). Puerarin evidently enhanced cultured Schwann cell viability, increased the density of myelinated axons, action potential area, and nerve conductive velocity, and consequently promoted nerve growth, which might be a potential medication for recovery of regenerating peripheral nerves (Hsiang et al, 2011).…”

“…The PR drug used in previous studies was referenced in this study. 39,40 In the PR-treated groups, PR was administered at a concentration of 50 mg/kg. The dose of PR is based on previous study in the experimental spinal cord injury.…”

“…Indeed, besides the anti-hyperglycemia and b-cell protection effects, puerarin was shown to have beneficial actions in ameliorating diabetic complications, including diabetic retinopathy (Teng et al 2009, Hao et al 2012, nephropathy (Shen et al 2009), vascular endothelial cell damage induced by high-glucose , and diabetic osteoporosis . In addition, puerarin is also prescribed for cardiovascular disease in China, and could accelerate peripheral nerve regeneration, and thus might be beneficial to diabetic neuropathy (Hsiang et al 2011) as well. With those pleiotropic benefits on insulin-producing b-cell and its target tissues concomitantly, puerarin may serve as a therapeutic option for diabetes and its complications.…”
