Purification and characterization of five different α subunits of guanine‐nucleotide‐binding proteins in bovine brain membranes

Abstract: We have purified five different a subunits of guanine-nucleotide-binding proteins (G proteins) from bovine brain membranes as active forms bound to guanosine 5'-[y-thio]triphosphate (GTP[yS]). All the purified a subunits were interacted with py subunits and served as a substrate for pertussin-catalyzed ADP-ribosylation. Based on the findings of immunoblot analyses using specific antibodies raised against various CI subunits of G proteins, three of them were identified as M~-~, M~-~ and M~-~, and the other two … Show more

“…When AlF 4 Ϫ was applied, a mixture of 100 M AlCl 3 (Sigma-Aldrich) and 10 mM NaF (Sigma-Aldrich) was added to the solution. GST-RGS4, CaM (Sigma-Aldrich), and purified G ␤␥ subunit from bovine brain (19) were diluted into experimental solutions just before use. Phosphatidylinositol-3,4,5-trisphosphate (PIP 3 ), phosphatidylinositol-4,5-bisphosphate (PIP 2 ), phosphatidylinositol-4-phosphate (PIP) were purchased from Biomol Research Laboratories (Plymouth Meeting, PA), and phosphatidylinositol (PI) was purchased from Sigma-Aldrich.…”

“…Finally, contradictory data are available in the literature about the ability of G o to inhibit adenylyl cyclase (Kobayashi et al, 1990;Wong et al, 1992). Our results first suggest that Ga oA is able to mediate inhibition of adenylyl cyclase activity in GHsecreting adenoma cells after SST5 activation, but its role is not essential for this effects, since it was maintained after Ga oA silencing.…”
