Purification, Characterization, and Chemical Modification of Neurotoxic Peptide from Daboia russelii Snake Venom of India

Abstract: Comprehensive knowledge of venom composition is very important for effective management of snake envenomation and antivenom preparation. Daboia russelii venom from the eastern region of India is the most neurotoxic among the four venom samples investigated. From the eastern D. russelii venom sample, neurotoxic peptide has been purified by combined method of ion exchange gel permeation chromatography and reversed phase high performance liquid chromatography. Molecular weight of Daboia neurotoxin III (DNTx-III) … Show more

“…Tandem mass spectrometry of venom fractions (n = 10) from these samples identified between 49 and 66 proteins from 13 toxin families, including phospholipase A 2 (PLA 2 ), Kunitz-type serine protease inhibitor (Kunitz), snake venom serine protease (SVSP), cysteinerich secretory protein (CRISP), snaclec, snake venom metalloproteinase (SVMP), three-finger toxin [3FTx; neurotoxic-3FTx (N-3FTx) and cytotoxic-3FTx (C-3FTx)], L-amino-acid oxidase ). The identification of 3FTx in Daboia venoms is particularly noteworthy as these toxins are rarely reported in the venom proteomes of viperid snakes [36,37].…”

“…Although significant research has focused on the neurotoxic effects of venoms, comparatively little work has been done on the coagulotoxic effects of snake species due to the inherent difficulties in working with blood without the use of advanced robotics such as that utilized in this study [15,16,17]. Venoms acting upon the blood are often utilized by species which predate upon mammals, effectively disrupting hemostasis within the body leading to hemorrhagic shock or stroke [18,19].…”

“…A number of studies had been made on snake venoms and their isolated protein components for investigating the neurotoxic and myotoxic effects [2432]. It was well-known that not only the neurotoxins and the myotoxins were extensively discovered as the main components in the venoms of Elapidae as well as Hydrophiidea, but also recently they were found as the minor components in the venoms of Viperidae and Crotalidae, especially Colubridae.…”
