Pyroptosis, a New Breakthrough in Cancer Treatment

Abstract: The way of cell death can be roughly divided into two categories: cell necrosis and PCD(programmed cell death). Pyroptosis is a kind of PCD, its occurrence depends on the gasdermin protein family and it will produce inflammatory response. With constant research in recent years, more and more evidences show that pyroptosis is closely related to the occurrence and development of tumors. The treatment of tumors is a big problem worldwide. We focus on whether we can discover new potential tumor markers and new the… Show more

“…Deregulation of apoptosis is considered one of the characteristics of cancer progression, and transformed cells are able to circumvent this process, although the mechanisms involved are not sufficiently known. For this reason, therapeutic strategies aimed at restoring the sensitivity of cancer cells to apoptosis are increasingly tested [113][114][115]. Citrus fruits represent major sources of flavonoids.…”

“…In addition, oleuropein induced the downregulation of metalloproteinase genes, which may be involved in the prevention of breast cancer metastases [113,116,181]. An antioxidant and growth inhibitory effect was found for differentiated thyroid cancer [115].…”

“…We obtained 52 pyroptosis-related genes (PRGs) from prior reviews (Xia et al, 2019;Zhou and Fang, 2019;Li et al, 2020;Ju X. et al, 2021;Wu et al, 2021;Ye et al, 2021) and MSigDB database v7.4 (Subramanian et al, 2005) (listed in Supplementary Table S1). We identified DEPRGs between tumor and normal tissues using the "limma" package, with a p-value < 0.05.…”

“…Pyroptosis is a newly discovered form of programmed cell death that is morphologically distinct from apoptosis and necrosis while releasing inflammatory mediators in the process (Wu et al, 2021). Pyroptosis is mediated by pore-forming proteins, such as the gasdermin family, of which gasdermin D (GSDMD) is a primary substrate for the caspase family (Li L. et al, 2021).…”

“…Pyroptosis is a programmed cell death process that was first described when primary mouse macrophages were treated with an anthrax lethal toxin [ 1 , 2 , 3 , 4 , 5 , 6 ]. It was mistakenly reported as apoptosis when the macrophages were infected with Shigella flexneri in 1992 [ 7 ].…”
