Hepatic ischemia reperfusion (IR) is an inevitable clinical problem for surgical procedures such as liver transplantation and liver resection. This study was designed to evaluate the protective effect of perindopril (PER) against hepatic IR injury. Thirty‐two rats were used and randomly allocated into four groups. Sham control group was subjected to sham operation and received saline only, IR group was subjected to IR and received vehicle, PER group was pretreated with PER (one milligram per kilogram per day i.p. for 10 consecutive days), and IR+PER group was pretreated with PER then subjected to IR. Liver function biomarkers (aspartate aminotransferase and alanine aminotransferase), oxidative stress (glutathione, malondialdehyde, myeloperoxidase, and superoxide dismutase) and inflammation markers (tumor necrosis factor‐alpha, interferon‐gamma, and inteleukin‐10 [IL‐10]), mRNA expression of NF‐κB‐p65 and TLR‐4, as well as protein expression of JAK1, STAT‐3, PI3K, mTOR, Akt, and Nrf‐2 were investigated concomitantly with histopathological examination. The results indicated that, hepatic IR induced a significant alteration in liver function biomarkers and structure, oxidative stress, and inflammation. At the molecular level, up‐regulation of NF‐κB‐p65, TLR‐4, JAK1, and STAT‐3 concomitantly with down‐regulation of Nrf‐2, IL‐10, PI3K, Akt, and mTOR were observed. These disturbances were alleviated by pretreatment of IR rats with PER in concomitant with hepatic structural improvement. Conclusively, the protective effect of PER presumably may be relevant to its ability to reduce oxidative stress, ameliorate the inflammatory processes, and modify the related signaling pathways. Background: Liver ischemia reperfusion (I/R) damage which is frequently seen in clinical hepatobiliary surgeries has no effective treatment for it. Liv-52, known to have hepatoprotective effects, is a natural antioxidant drug licensed by the Ministry of Health of India. The aim of our study is to investigate the effect of Liv-52 on liver damage induced by I/R in rats. Methods: Albino Wistar male rats were divided into three groups; liver I/R (IR), 20 mg/kg Liv-52 + liver ischemia reperfusion (LIR) and sham operation applied to control group (HG). Liv-52 was administered to the LIR group (n = 6) 1 h prior to I/R application and distilled water was given orally to IR (n = 6) and HG (n = 6) groups as a solvent. Ischemia was determined as 1 h, and reperfusion was identified as 6 h in animals. Results: Increased levels of alanine aminotransferase, aspartate aminotransferase and lactate dehydrogenase, malondialdehyde, myeloperoxidase, and decreased levels of superoxide dismutase, and glutathione related enzymes caused by I/R application have been converged to healthy group level with Liv-52 treatment and the damage in liver tissue has been improved histopathologically. Conclusions: Liv-52 may be beneficial for preventing liver I/R damage in pre-surgery application.

Objective: In burn injury, the zone of stasis determines the width and depth of the necrosis. Our aim is to show the effectiveness of quercetin on the viability within the zone of stasis in burns of rats. Materials and Methods: Forty-eight rats were divided into three groups. The rats in Group 1 (control group) were only applied the comb burn model; the rats in Group 2 (post-burn group) were administered 50 mg/kg of quercetin intraperitoneally, every day after the burn procedure until euthanasia; and the rats in Group 3 (pre-burn group) were administered 50 mg/kg of quercetin intraperitoneally, every day for 7 days before and after the burn procedure until euthanasia. Results: The living tissue calculated was 85.41% (±14.06) in Group 3, 40.37% (±9.75) in Group 2, and 16.81% (±9.4) in Group 1. The level of apoptosis was 30.0 (±10.8) in Group 3, 33.8 (±08.7) in Group 2, and 37.4 (±11.5) in Group 1. The level of autophagy was 49.50 (±8.58) in Group 3, 27.17 (±5.53) in Group 2, and 21.00 (±5.66) in Group 1. All the differences between the groups were statistically significant (p<0.01). Conclusion: Quercetin reduces apoptosis and increases autophagy, thereby increasing tissue viability in the zone of stasis of burn injury.
