Quinoline, with the active site of 8‐hydroxyl, efficiently inhibits Micropterus salmoides rhabdovirus (MSRV) infection in vitro and in vivo

Abstract: Micropterus salmoides rhabdovirus (MSRV) is an significant pathogen that causes high mortality and related economic losses in bass aquaculture. There is no effective or approved therapy to date. In this study, we evaluated the anti-MSRV effects of 22 quinoline derivatives in grass carp ovary (GCO) cells. Among these compounds, 8-hydroxyquinoline exhibited valid inhibition in decreasing MSRV nucleoprotein gene expression levels of 99.3% with a half-maximal inhibitory concentrations (IC 50 ) value of 4.66 μM at … Show more

“…In a separate study by Li et al, the anti‐MSRV effects of 22 quinoline derivatives were evaluated in GCO cells. Notably, 8‐hydroxyquinoline exhibited strong inhibition with an IC 50 value of 4.66 μM at 48 h, resulting in a 99.3% decrease in MSRV nucleoprotein gene expression levels 79 . Moreover, ursolic acid was found to inhibit MSRV replication in GCO cells with an IC 50 value of 5.55 μM 80 …”

“…Notably, 8-hydroxyquinoline exhibited strong inhibition with an IC 50 value of 4.66 μM at 48 h, resulting in a 99.3% decrease in MSRV nucleoprotein gene expression levels. 79 Moreover, ursolic acid was found to inhibit MSRV replication in GCO cells with an IC 50 value of 5.55 μM. 80 MSRV is highly contagious and can spread rapidly among bass populations through water and contaminated equipment, which can result in significant economic losses in the aquaculture industry.…”

“…8-Hydroxyquinoline derivatives have been reported to have wide biological activities, such as anticancer activity, antiviral activity, antibacterial activity, and antifungal activity . Two well-known members of this family, clioquinol and nitroxoline are broad-spectrum antimicrobial agents (Figure ).…”

“…The DNA vaccine and subunit vaccine prepared with glycoprotein of MSRV had good immune protection effects against MSRV infection (Guo et al., 2020; Xu et al., 2022; Yang, Guo, et al., 2022; Yang, Xu, et al., 2022). In addition, Chinese herbal medicine (magnolol derivatives) and antiviral drug (ribavirin and 8‐hydroxyquinoline) could decrease tissue viral nucleic acids, and improve the survival rate of largemouth bass after MSRV infection (Jin et al., 2022; Li et al., 2022; Yang et al., 2021). While the efficacy and mechanism of disinfectants against MSRV in water are still lacking.…”
