Radioimmunoassay for Desmosine

Abstract: A radioimmunoassay was developed for the determination of desmosine. Desmosine conjugated to albumin was injected into rabbits which developed useful titers of serum antibodies after six months. A radioactive probe was prepared with desmosine using [125I]-Bolton-Hunter reagent. Bound desmosine was separated from free desmosine by cellulose acetate filter binding. The sensitivity of the assay is 1-50 picomoles of desmosine. The antibody is highly selective for desmosine, reacting less than 1% with other known c… Show more

“…2). Immunology-based methods for DES detection in urine include ELISA [11] and radioimmunoassay (RIA) [12,13]. The latter method has undergone progressive modifications, after which urine did not require acid hydrolysis [14], and the specificity improved [6].…”

“…Early work, in which urinary DES was detected by RIA [12], compared a group of both children and adults (the latter with or without emphysema) who had AATD (PI*ZZ genotype) with appropriate controls. Urinary DES in children (mean age 10 yrs) was significantly higher than it was in all adults (mean¡SD 3.50¡0.62 versus 2.32¡0.82 mg per 100 mg creatinine, respectively; p ,0.001), but levels in children with AATD were not different from those of age-matched controls [28].…”

“…Because desmosine is unique to elastin in mammals (it is, however, found in avian and reptilian egg-shell membranes [26,27]), it can be used as a quantitative marker for insoluble elastin levels in tissues and cultured cells. Many methods have been employed for detection and quantification of desmosine, including HPLC [28][29][30][31][32][33], radioimmunoassay [27,[34][35][36][37], ELISA [38][39][40], capillary electrophoresis, [41] and mass spectrometry [42][43][44]. Of these techniques, only RIA and ELISA are applicable to high throughput analysis.…”

“…While the RIA provides reproducible results and the specificity of the antibody for desmosine has been documented in both urine and lung homogenates, 19,33 recent methodological advances 34 in liquid chromatography and tandem mass spectrometry may allow for more sensitive and specific measurements of desmosine for future studies.…”
