Changes in the levels of serum steroids and ovarian steroidogenic enzyme mRNA were analyzed in Japanese eel Anguilla japonica induced to undergo oogenesis by chum salmon pituitary homogenate (SPH) treatment. Serum 17α‐hydroxyprogesterone, 17α,20β‐dihydroxy‐4‐pregnen‐3‐one, androstenedione and estrone levels were barely or not detectable during artificial maturation. In contrast, serum estradiol‐17β (E2) and testosterone (T) levels increased after SPH injections and peaked at the migratory nucleus (MN) stage. 3β‐Hydroxysteroid dehydrogenase mRNA levels did not change between eels in different stages of oogenesis. mRNA levels of P450 cholesterol side‐chain cleavage (P450scc), P450 17α‐hydroxylase/C17‐20 lyase (P450c17), 17β‐hydroxysteroid dehydrogenase type I (17β‐HSD‐I) and P450 aromatase (P450arom) were low before SPH injections. P450scc, P450c17 and 17β‐HSD‐I transcript abundance increased after SPH treatment and peaked at the MN stage. P450arom mRNA levels, however, peaked at the mid‐vitellogenic stage and slightly decreased towards the MN stage. We suggest that these increases in P450scc and P450c17 mRNA levels may account for high T levels at the MN stage. In turn, the high T levels may permit the production of E2 in spite of low P450arom mRNA levels towards the end of oogenesis. Steroidogenesis in artificially maturing eels appears to proceed unlike that in other teleosts, but whether or not this is an artifact remains as yet unknown.

To harmonise vertebrate OECD Test Guidelines for endocrine disruption testing between mammalian and non-mammalian test species, additional Estrogen, Androgen, Thyroid and Steroidogenesis (EATS) modality endpoints in non-mammalian models need to be assessed. These would mean for example the addition of hormonal measurements in fish, birds and amphibians. Furthermore, a better reporting of gross pathology findings for birds would also be considered advantageous for the assessment of endocrine disrupting properties. To facilitate adoption of additional measures, guidance on how to perform, report and evaluate these new endpoints is required. In this report, a variety of methods including a systematic evidence map, an extensive literature review and a survey of ecotoxicology laboratories were adopted to collect data on the topic. The systematic evidence map uncovered a range of methods for measuring sex and thyroid hormones in fish, birds and amphibians, although methods for measuring sex-hormones in fish, were by far, the most frequently encountered in the literature and laboratory survey. However, there are still considerable gaps in knowledge for: optimum sample timing for hormonal measurement (diurnal, developmental stage, etc.), issues with inherent variability, low sample volume (plasma/serum), test species selection, possible impacts of housing/diet/stress. The extensive literature review revealed that although gross pathology and histopathology have been used to investigate the effects of endocrine disrupting chemicals in birds, there are no standardised methods for assessment or interpretation, although relative weight of endocrine organs is frequently used as a gross pathology metric. Reports of how histopathology was assessed varied considerably. From the survey, few contract laboratories are experienced in conducting the avian test guideline and these types of pathology techniques. Recommendations for future work with non-mammalian taxa include: investigating the optimal time (or timings) for measuring hormones, developing non-invasive hormone measuring techniques, gaining knowledge of baseline/control hormonal data, and further developing guidance on conducting and assessing gross pathology and histopathology in birds.

VINITHKUMAR GOPINATHAPILLAI SREERAJ MUTHUKAMATCHI RAVINDRAN Abstract Vertebrate-type steroids (estradiol-17β and progesterone) and 5-hydroxytryptamine (5-HT) were examined during the different stages of ovarian maturation in the Indian spiny lobster Panulirus homarus. Estradiol-17β and progesterone in the haemolymph and ovary were quantified by radioimmunoassay. Estradiol-17β was not detectable in the haemolymph when the oocytes were in stage I. It appeared in the haemolymph only as the oocytes attained stage II. Subsequently, a sharp increase in the level of estradiol-17β was observed in the haemolymph of lobsters with stage III oocytes; it then showed a significant fall when the ovary was full of stage IV oocytes. Although progesterone was also not detectable in the haemolymph when the oocytes were in stage I, it gradually increased during stages II and III and reached a peak level during stage IV. Surprisingly, both estradiol-17β and progesterone were detectable in the ovary from stage I onwards. In the ovary, estradiol-17β and progesterone showed peak levels during stages III and IV, respectively. A stage-dependent change in the activity and distribution of 5-hydroxytryptaminergic neurons in the brain and thoracic ganglia was also M04013; observed immunocytochemically in relation to ovarian recrudescence. Furthermore, HPLC-EC conducted on the level of 5-HT in the brain and thoracic ganglia indicated similar changes in relation to ovarian maturation. These results strongly suggest that 5-HT is involved in ovarian development through certain inhibitory/stimulatory factors present in the X-organ-sinus gland complex of the eye-stalk in the spiny lobster P. homarus.
